TRIP13 knockdown inhibits the proliferation, migration, invasion, and promotes apoptosis by suppressing PI3K/AKT signaling pathway in U2OS cells

De-Chen Yu et al.

Summary: This study used bioinformatics strategies to identify key genes associated with osteosarcoma (OS) and validated the role of TRIP13. Knockdown of TRIP13 can inhibit the proliferation, migration, and invasion of U2OS cells by downregulating the PI3K/AKT signaling pathway, providing new insights for the early diagnosis and treatment of osteosarcoma.

TGR5 deficiency activates antitumor immunity in non-small cell lung cancer via restraining M2 macrophage polarization

Lifang Zhao et al.

Summary: This study reveals the protumor function of TGR5 in non-small cell lung cancer (NSCLC) by regulating tumor-associated macrophage (TAM) polarization and suppressing anti-tumor immunity. Mechanistically, TGR5 promotes the transformation of TAMs into protumorigenic M2-like phenotypes through the activation of the cAMP-STAT3/STAT6 signaling pathway. The study also shows that the expression of TGR5 is associated with TAM infiltration in NSCLC tissues, and the co-expression of TGR5 and high TAM infiltration correlates with the prognosis and overall survival of NSCLC patients.

Cadmium induces epithelial-mesenchymal transition and migration of renal cancer cells by increasing PGE2 through a cAMP/PKA-COX2 dependent mechanism

Haifeng Shi et al.

Summary: This study found that low-dose Cd exposure promoted migration of renal cancer Caki-1 cells, which was not dependent on Cd-induced ROS and intracellular Ca2+ levels. Cd exposure induced cAMP/PKA-COX2 signaling pathway, mediating cell migration and invasion, and decreasing expression of EMT marker E-cadherin while increasing expressions of N-cadherin and Vimentin. Moreover, Cd-induced secretion of PGE2 feedback on activation of cAMP/PKA-COX2 signaling, also promoting EMT, migration, and invasion of renal cancer Caki-1 cells.

cAMP levels regulate macrophage alternative activation marker expression

Swamy Polumuri et al.

Summary: The capacity for macrophages to polarize into distinct functional activation states is critical for tuning inflammatory responses. In addition to IL-4/IL-13, signals from the inflammatory milieu can further modulate M2 marker expression, with cAMP playing a role in increasing M2 markers through a PKA/C/EBP beta/CREB dependent pathway.

Pan-cancer analysis and experiments with cell lines reveal that the slightly elevated expression of DLGAP5 is involved in clear cell renal cell carcinoma progression

Yun Feng et al.

Summary: DLGAP5 is upregulated in various cancers, including ccRCC, and its expression is positively correlated with immune infiltration levels in ccRCC. Elevated DLGAP5 expression is associated with ccRCC progression and may serve as a prognostic risk factor, but not an independent one.

Laboratory screening test with inhalant and food allergens in atopic Brazilian children and adolescents: a performance

Ana Carolina Rozalem Reali et al.

Summary: Phadiatop Infant (R) is a useful tool to assess allergic sensitization in children under 5 years old. A study in Brazilian children showed high positivity of the test, especially in cases of food allergy and atopic dermatitis. The test had high sensitivity and specificity compared to specific serum IgE dosage, making it a valuable screening method for allergic diseases in non-European countries.

Epalrestat suppresses inflammatory response in lipopolysaccharide-stimulated RAW264.7 cells

Keisuke Sato et al.

Summary: This study demonstrated that EPS suppresses inflammatory response in LPS-stimulated RAW264.7 cells by regulating the levels of pro-inflammatory cytokines and inflammatory mediators. This suggests that targeting the regulation of these molecules by EPS could be a promising therapeutic approach for treating inflammatory injury. EPS, with confirmed safety and pharmacokinetics clinically, is expected to be useful for the treatment of inflammatory diseases.

Hui Wang et al.

Summary: Colorectal cancer (CRC) is a highly prevalent and lethal malignant tumor in the digestive system. Tumor-associated macrophages (TAMs) play a crucial role in various mechanisms of CRC progression, such as promoting tumor proliferation, invasion, angiogenesis, and immunosuppression, although their exact role is still debated in clinical evidence.

Summary: PLXDC2 is a gene associated with the tumor microenvironment, linked to poor prognosis and advanced disease in gastric cancer patients, and correlated with the expression of M2 macrophages and inflammatory signaling pathways. Its expression in stromal cells and interaction with tumor-associated macrophages may induce the EMT process, contributing significantly to cancer biology.
