Summary: In hepatocellular carcinoma (HCC), PD-L1(+) host macrophages are a major cellular source of PD-L1 in tumors, displaying a glycolytic phenotype and producing antitumor IL-12p70. The regulation of these macrophages involves fibronectin 1-induced PKM2, controlling both antitumor properties and inflammation-mediated PD-L1 expression. Modulating the context of glycolytic macrophages in HCC tumors may restore their antitumor properties and offer a precise strategy for anticancer therapy.

Hyperthermia promotes exosome secretion by regulating Rab7b while increasing drug sensitivity in adriamycin-resistant breast cancer

Summary: This study investigated the mechanism through which hyperthermia promotes exosome secretion and drug sensitivity in adriamycin-resistant breast cancer. The results showed that hyperthermia suppressed the growth of cancer cells and increased drug sensitivity. It also upregulated certain genes related to longer overall survival in breast cancer patients. Hyperthermia promoted exosome secretion through a small GTPase called Rab7b. These hyperthermia-triggered exosomes can carry antitumor genes to other cells and may enhance the effects of hyperthermia. This study provides new insights into the relationship between therapeutic strategies and exosome biology.

The miR-21-5p and miR-200a derived from sEVs play key regulatory roles in this process. Inhibiting the secretion of specific sEV-miRNAs from CRC and targeting PD-L1 in TAMs may serve as novel methods for CRC treatment.

Role of Biological Mediators of Tumor-Associated Macrophages in Breast Cancer Progression

Summary: This review highlights the biological mediators associated with tumor-associated macrophages (TAMs) in the tumor microenvironment (TME) of breast cancer (BRCA), including cancer-associated fibroblasts, endothelial cells, adipocytes, tumor-derived exosomes, extracellular matrix, and other immune cells. Increased infiltration of TAMs and elevated expression of TAM-related genes have been linked to a poor prognosis of BRCA. TAM-targeted therapeutic strategies, such as inhibitors of CSF-1/CSF-1R, CCL2/CCR2, CCL5-CCR5, bisphosphonate, nanoparticle, and exosomal-targeted delivery, have been developed and are currently being tested in intervention trials.

Summary: This study reveals that circPSMA1 functions as a tumor promoter in triple-negative breast cancer (TNBC) by acting as a miRNA sponge to absorb miR-637 and regulate Akt1-beta-catenin (cyclin D1) axis. Exosomes from TNBC cells carrying high levels of circPSMA1 can transmit migration and proliferation capacity to recipient cells. High expression of Akt1 and low expression of miR-637 are highly correlated with poor prognosis in TNBC patients with lymph node metastasis.
