APC/C CDH1 ubiquitinates STAT3 in mitosis

Summary: STAT3, an oncogene associated with poor prognosis, can be ubiquitinated by APC/C CDH1 during mitosis, resulting in long-lasting anti-tumor responses. Inhibiting phosphatases can decrease STAT3 ubiquitination. Targeting mitosis may be a potential therapeutic window for treating STAT3-activated cancers.

CAFs secrete CXCL12 to accelerate the progression and cisplatin resistance of colorectal cancer through promoting M2 polarization of macrophages

Summary: The study aimed to investigate the impact of the interaction between tumor-associated fibroblasts (CAFs) and macrophage polarization on colorectal cancer (CRC) progression. Clinical tissue samples of CRC were collected to isolate normal fibroblasts (NFs) and CAFs. Co-culture of colorectal cancer cell lines with CAFs resulted in increased proliferation, migration, invasion, and cisplatin (DDP) resistance. CAFs exhibited a higher ability to recruit macrophages and induce M2 polarization through the secretion of CXCL12. In vivo experiments confirmed that CAFs promoted CRC progression and DDP resistance by affecting M2 polarization mediated by CXCL12.

KDM2A induces p53-dependent senescence and promotes the release of cytokines in fibroblasts, thereby promoting cancer cell proliferation. Summary: This study confirmed the cancer-promoting roles of CAFs in bladder cancer, with CALD1 identified as a key gene associated with CAFs that may promote bladder cancer progression by remodeling the tumor microenvironment. Bioinformatics methods such as CIBERSORT, MCP-COUNTER, and ESTIMATE algorithms provide important value for studying the tumor microenvironment, showing significant correlations between CALD1 and various components. Immunohistochemistry results validated the strong association of CALD1 with CAFs and macrophages.
