Haematopoietic cell transplantation for children with acute megakaryoblastic leukaemia without Down syndrome

Asahito Hama et al.

Summary: This study retrospectively analyzed the outcomes of hematopoietic cell transplantation (HCT) in 203 children with non-DS-AMKL. The results showed that the 5-year overall survival rate was higher in patients who underwent HCT in CR1 and from an HLA-matched donor. Risk stratification should be established for non-DS-AMKL patients to determine HCT indication in CR1.

Unmanipulated haploidentical hematopoietic stem cell transplantation for pediatric de novo acute megakaryoblastic leukemia without Down syndrome in China: A single-center study

Junbin Huang et al.

Summary: A retrospective study on 25 pediatric AMKL patients without DS was conducted, and the results showed that patients with trisomy 19 and MRD negative pre-HSCT had better outcomes. Therefore, haploidentical HSCT may be an option for high-risk AMKL patients without DS.

Pediatric non-Down's syndrome acute megakaryoblastic leukemia patients in China: A single center's real-world analysis

Aoli Zhang et al.

Summary: This retrospective study investigated the clinical impact and prognosis of newly diagnosed non-Down's syndrome acute megakaryocytic leukemia (non-DS-AMKL) in children. The results showed that intensive chemotherapy improved the prognosis of non-DS-AMKL children. Subclassification based on bone marrow aspiration and immunophenotypic was proposed. However, the prognosis of non-DS-AMKL children remains unsatisfactory compared to other types of acute myeloid leukemia (AML).

Hematopoietic Cell Transplantation in the Treatment of Pediatric Acute Myelogenous Leukemia and Myelodysplastic Syndromes: Guidelines from the American Society of Transplantation and Cellular Therapy

Katherine Tarlock et al.

Summary: This evidence-based review evaluates the role of allogeneic hematopoietic stem cell transplantation (HCT) in treating acute myelogenous leukemia (AML) in children. Genomic characterization and treatment response are important for risk stratification in pediatric AML. Allogeneic HCT offers survival benefits for selected patients with unfavorable risk features and is standard care for children who relapse after initial chemotherapy.

Summary: This study compared the outcomes of different donor sources in AML patients undergoing transplantation and concluded that HID-HSCT had similar survival and relapse rates compared to MUD and MSD-HSCT, indicating that HID is a valid option.

Haploidentical Stem Cell Transplantation for Acute Myeloid Leukemia: Current Therapies, Challenges and Future Prospective

Ying-Jun Chang et al.

Summary: Haploidentical stem cell transplantation is an effective alternative donor source for AML patients, with outcomes comparable to traditional transplantation methods for various types of AML. Relapse and infections post haplo-SCT remain key challenges in improving clinical outcomes for AML patients. Areas of active research include relapse prophylaxis, intervention, treatment, as well as infection prevention and therapy.

Guan-Hua Hu et al.

Summary: The study assessed the efficacy and safety of bridging CAR-T therapy to Haplo-HSCT and found that Haplo-HSCT may be a safe and effective treatment strategy to improve event-free survival and overall survival after CAR-T therapy.
