New roles for AP-1/JUNB in cell cycle control and tumorigenic cell invasion via regulation of cyclin E1 and TGF-β2

Beatriz Perez-Benavente et al.

Summary: The transcription factor JUNB plays a role in cell division and tumorigenic processes by regulating the expression of cyclin El and transforming growth factor (TGF)-beta 2. High levels of JUNB can switch the response of TGF-beta 2 stimulation from anti-proliferative to pro-invasive. JUNB amplification is associated with poor prognosis in breast and ovarian cancer patients.

Cancer cells escape autophagy inhibition via NRF2-induced macropinocytosis

Hua Su et al.

Summary: Although many cancers rely on autophagy for nutrient acquisition, blocking autophagy alone does not effectively inhibit tumor growth. Research has shown that blocking autophagy can lead to pancreatic ductal adenocarcinoma (PDAC) upregulating an alternative nutrient pathway, macropinocytosis (MP), allowing tumor cells to extract nutrients from outside sources. This switch from autophagy to MP, driven by the activation of transcription factor NRF2, presents a potential therapeutic strategy for inhibiting tumor growth and regression.

Loss of activating transcription factor 3 prevents KRAS-mediated pancreatic cancer

Summary: The study suggests a critical role of ATF3 in pancreatic cancer development through promoting ADM and PanIN formation. Loss of ATF3 can reduce response to pancreatic injury and decrease the incidence of PDAC.

CCL28 Downregulation Attenuates Pancreatic Cancer Progression Through Tumor Cell-Intrinsic and -Extrinsic Mechanisms

Jingjing Yan et al.

Summary: CCL28 has been reported to be upregulated in pancreatic ductal adenocarcinoma (PDAC) patients and negatively correlated with overall survival. Knockdown of CCL28 inhibits proliferation of pancreatic cancer cells and deficiency of CCL28 attenuates tumor growth in mouse models by remodeling the tumor microenvironment.

TNF-α-producing macrophages determine subtype identity and prognosis via AP1 enhancer reprogramming in pancreatic cancer

Mengyu Tu et al.

Summary: Large-scale genomic profiling of pancreatic cancer has identified two distinct subtypes - 'classical' and 'basal-like' - within the stromal immune microenvironment, influencing differential prognosis. Basal-like neoplastic state is sustained through BRD4-mediated cJUN/AP1 expression, inducing CCL2 to recruit TNF-alpha-secreting macrophages, which in turn forces classical neoplastic cells into an aggressive state via lineage reprogramming. Integration of various data sources revealed JUNB/AP1 (classical) and cJUN/AP1 (basal-like) driven regulation of PDAC subtypes.

FOSL2 promotes VEGF-independent angiogenesis by transcriptionnally activating Wnt5a in breast cancer-associated fibroblasts

Xueying Wan et al.

Summary: The FOSL2/Wnt5a signaling axis plays a crucial role in breast cancer angiogenesis in a VEGF-independent manner, and targeting this axis in cancer-associated fibroblasts may offer a potential option for anti-angiogenesis therapy.
