Application of Marine Natural Products against Alzheimer's Disease: Past, Present and Future

Summary: Alzheimer's disease (AD), a neurodegenerative disease, affects the elderly and manifests as impairments in memory, language, cognition, visuospatial skills, and executive function. Current drugs slow down AD progression and relieve symptoms but cannot cure the disease. The pathogenesis of AD is unclear, but it is associated with beta-amyloid expression. Marine natural products have shown promise as potential drug candidates, with polysaccharides, carotenoids, polyphenols, sterols, and alkaloids presenting neuroprotective effects. Research is ongoing to discover novel and efficacious drug compounds for AD.

Astaxanthin as a King of Ketocarotenoids: Structure, Synthesis, Accumulation, Bioavailability and Antioxidant Properties

Summary: Natural astaxanthin is richer than synthetic astaxanthin, but cultivating it in photobioreactors is expensive and downstream processing techniques for solubilization are not cost-effective. As a result, the cost of astaxanthin is high, leading pharmaceutical and nutraceutical companies to opt for synthetic astaxanthin. This review discusses the chemical character of astaxanthin, more cost-effective cultivating techniques, and its bioavailability. Moreover, the antioxidant properties of this microalgal product against various diseases are discussed, which make it a promising drug for reducing inflammation and its consequences.

Tat-GSTpi Inhibits Dopaminergic Cells against MPP+-Induced Cellular Damage via the Reduction of Oxidative Stress and MAPK Activation

Summary: This study reveals the important role of glutathione S-transferase pi (GSTpi) in cellular processes such as anti-oxidation and signal transduction. The use of Tat-GSTpi fusion protein shows its ability to reduce DNA damage and reactive oxygen species (ROS) generation, and improve cell viability by regulating MAPKs, Bcl-2, and Bax signaling. Furthermore, the delivery of Tat-GSTpi protein into the brain of PD mouse model protects against dopaminergic neuronal cell death induced by MPTP.

Summary: This study evaluated the redox state of the typical 2-CysPrx subgroup in a Parkinson's disease model induced by PQ. PQ-induced ROS compartmentalization was observed in different organelles, which was reflected by the hyperoxidation pattern of 2-Cys Prdx detected by redox eastern blotting. The overexpression of Prdx5 effectively decreased PQ-mediated ROS levels and protected dopaminergic cells from cell death, making it a potential therapeutic target for PD.
