Apolipoprotein E ε4 disrupts oligodendrocyte differentiation by interfering with astrocyte-derived lipid transport

Kingston King-Shi Mok et al.

Summary: Carriers of the APOE4 variant are more susceptible to Alzheimer's disease due to altered lipid and cholesterol transport capabilities. This study examines the impact of APOE4 on oligodendrocytes and finds a disruption in myelination, possibly contributing to the development of Alzheimer's disease.

Total Cholesterol and APOE-Related Risk for Alzheimer's Disease in the Alzheimer's Disease Neuroimaging Initiative

Michelle M. Dunk et al.

Summary: This study investigates the relationship between total cholesterol and APOE-related AD risk. The results show that higher total cholesterol may be a significant contributor to AD risk, particularly in APOE4 carriers. These findings highlight a possible mechanism by which APOE confers AD risk and indicate the potential for AD risk modification through maintenance of healthy total cholesterol levels.

Apolipoprotein E and Alzheimer's Disease: Findings, Hypotheses, and Potential Mechanisms

Summary: Alzheimer's disease (AD) is a complex neurodegenerative disorder that is difficult to treat. Recent advancements in imaging technology and experimental techniques have allowed researchers to gain insights into the pathogenic mechanisms of AD. The major genetic risk factor for late-onset AD, apolipoprotein E4 (apoE4), plays a significant role in the initiation and progression of AD, although its exact mechanisms are not fully understood.

APOE Antibody Inhibits Aβ-Associated Tau Seeding and Spreading in a Mouse Model

Maud Gratuze et al.

Summary: The study evaluated the effects of the anti-human APOE antibody HAE-4 on amyloid plaques, A beta-mediated tau seeding/spreading, and neuritic dystrophy in 5XFAD amyloid mice. HAE-4 reduced A beta plaques, A beta-driven tau seeding/spreading, and neuritic dystrophy, suggesting therapeutic potential on A beta removal and downstream consequences such as tauopathy.

The detrimental effects of APOE4 on risk for Alzheimer's disease may result from altered dendritic spine density, synaptic proteins, and

Lisa R. Taxier et al.

Summary: The study found that APOE4 has a negative impact on object memory, dendritic spine density, and levels of hippocampal synaptic proteins and ER alpha. Male E3FAD mice exhibited the best memory in two object-based tasks. APOE4 reduced basal dendritic spine density in the medial prefrontal cortex and dorsal hippocampus. These findings provide new evidence for understanding the interactions between APOE4 and sex.

APOE4 impairs myelination via cholesterol dysregulation in oligodendrocytes

Joel W. Blanchard et al.

Summary: APOE4 has widespread effects on gene expression in the human brain, particularly in relation to cholesterol homeostasis and transport. Aberrant cholesterol deposition in oligodendrocytes leads to reduced myelination, while pharmacologically facilitating cholesterol transport improves learning and memory. This study provides insights into the impact of APOE4 on the aging human brain and suggests potential therapeutic strategies for Alzheimer's disease.

APOE4 drives inflammation in human astrocytes via TAGLN3 repression and NF-?B activation

Laurie Arnaud et al.

Summary: This study reveals that APOE ε4 controls inflammation in human astrocytes by regulating TAGLN3 expression and NF-KB activation. APOE4 downregulates TAGLN3, resulting in chronic low-grade inflammation and hyperactivated inflammatory responses. TAGLN3 supplementation can pharmacologically reverse this pro-inflammatory state, and TAGLN3 is confirmed to be downregulated in the brain of patients with sAD.

These biomarkers may hold promise for monitoring and predicting AD progression.

Eicosanoid lipidome activation in post-mortem brain tissues of individuals with APOE4 and Alzheimer's dementia

Brandon Ebright et al.

Summary: This study compared the lipidome in the dorsolateral prefrontal cortex of older individuals with AD dementia and age-matched controls with no cognitive impairment. The results showed lower levels of neuroprotectin D1 and omega-3 fatty acids in AD dementia patients, which were associated with lower cognitive function. Carrying the APOE4 allele was found to be associated with higher levels of pro-inflammatory and pro-resolving eicosanoid lipids. Furthermore, the activation of certain enzymes involved in lipid metabolism pathways was observed. These findings highlight the role of the eicosanoid lipidome in chronic inflammation in AD dementia and suggest potential therapeutic targets.

Hatice Eraslan Boz et al.

Summary: The study aimed to investigate prosaccade stimulus eccentricity and compare prosaccade parameters in patients with AD, aMCI, and NA. The results showed that patients with AD had lower correct saccade rates and longer latencies than NA. Patients with aMCI did not differ in prosaccade performance compared to NA. Prosaccade parameters were significantly correlated with all cognitive domains.

Apolipoprotein Mimetic Peptides: Potential New Therapies for Cardiovascular Diseases

Anna Wolska et al.

Summary: Since the breakthrough of using insulin to treat diabetic patients in the 1920s, there has been a growing interest in developing various proteins and their peptide mimetics for treating medical disorders. Currently, there are over 60 approved peptides for human use and more than 150 peptides in clinical development. This review mainly focuses on peptide mimetics related to cardiovascular diseases, discussing the biochemistry, design, and clinical trials of peptides based on apoA-I, apoE, and apoC-II, while also addressing the limitations and challenges in developing apolipoprotein mimetic peptides into new drugs.
