Plk1 regulates mutant IDH1 enzyme activity and mutant IDH2 ubiquitination in mitosis

M. Saikiran Reddy et al.

Summary: The study reveals that the mitotic kinase Plk1 regulates the function of mutant IDH1 and IDH2 in mitosis. Plk1 phosphorylates mutant IDH1 and IDH2, thereby controlling their enzyme activity and ubiquitination. These findings suggest that Plk1 could be a potential therapeutic target in mutant IDH-linked tumors.

Regulation of gene expression by glycolytic and gluconeogenic enzymes

Xueli Bian et al.

Summary: Gene transcription and cell metabolism are two interrelated biological processes, with altered glucose metabolic gene expression being a major driver of enhanced aerobic glycolysis in tumor cells. Notably, glycolytic and gluconeogenic enzymes in tumor cells also have additional functions and directly regulate gene expression by modulating chromatin or transcriptional complexes. The mutual regulation between cellular metabolism and gene expression is a unique feature of tumor cells and provides specific molecular and functional targets for cancer treatment.

Reconstruction of a catalogue of genome-scale metabolic models with enzymatic constraints using GECKO 2.0

Ivan Domenzain et al.

Summary: This study upgrades the GECKO toolbox to enhance models with enzyme and proteomics constraints for any organism with a compatible GEM reconstruction. Enzyme-constrained models for three different yeasts were generated and used to study their long-term adaptation to stress factors. Predictions suggest the importance of metabolic robustness in microbial growth under stress and nutrient-limited conditions.

Interrogating the effect of enzyme kinetics on metabolism using differentiable constraint-based models

St Elmo Wilken et al.

Summary: In this paper, the sensitivity analysis of constraint-based metabolic models is studied, and several applications of this technique are demonstrated. By calculating the sensitivity of reaction fluxes and enzyme concentrations to parameters, rate limiting enzymes can be identified and parameter estimation can be improved. Furthermore, this technique can be expanded to study complex models and validated against experimental data.

GeneReg: a constraint-based approach for design of feasible metabolic engineering strategies at the gene level

Zahra Razaghi-Moghadam et al.

Summary: Existing computational approaches often neglect the impact of gene expression regulation on metabolic engineering strategies. This study finds that the association of genes with multiple reactions may render engineering strategies infeasible at the flux level, and proposes a constraint-based approach called GeneReg to facilitate the design of feasible strategies at the gene level.

Glycolysis-induced drug resistance in tumors-A response to danger signals?

Fabrizio Marcucci et al.

Summary: Tumor cells often switch from mitochondrial oxidative metabolism to glycolytic metabolism, leading to drug resistance. Glycolysis-induced drug resistance may involve inhibition of apoptosis, induction of epithelial-mesenchymal transition, among other mechanisms.

A hexokinase isoenzyme switch in human liver cancer cells promotes lipogenesis and enhances innate immunity

Laure Perrin-Cocon et al.

Summary: During the transformation into hepatocellular carcinoma, the replacement of GCK by HK2 is associated with poor prognosis for patient survival. Knocking out HK2 and expressing GCK in Huh7 cell line rewired central carbon metabolism, stimulated mitochondrial respiration, and reactivated innate immune responses, showing that consequences of the HK switch extend beyond metabolic reprogramming.

Integrating systemic and molecular levels to infer key drivers sustaining metabolic adaptations

Pedro de Atauri et al.

Summary: This study develops an efficient computational strategy to infer key molecular drivers sustaining metabolic adaptations to complex perturbations by integrating measured changes at systemic and molecular levels and combining metabolic control analysis with linear programming tools. The reprogramming of transporter and enzyme activities is identified as essential for orchestrating the metabolic adaptation to antitumoral drug therapy in colon cancer cells.
