Pseudomonas umsongensis GO16 as a platform for the in vivo synthesis of short and medium chain length polyhydroxyalkanoate blends

β-oxidation-polyhydroxyalkanoates synthesis relationship in Pseudomonas putida KT2440 revisited

Summary: Through studying Pseudomonas putida KT2440, it was found that PhaJ4 is the main supplier of monomers for PHA synthesis, while PhaJ1 has preference for C6 substrate. Even when all PhaJ homologues were deleted, the mutant still accumulated PHA. Further proteome analysis revealed PedE and PedH as potential monomer suppliers, but even with their deletion, PHA levels remained unchanged. Therefore, PHA metabolism exhibits redundancy.

Genome-centric polyhydroxyalkanoate reconciliation reveals nutrient enriched growth dependent biosynthesis in Bacillus cereus IBA1

Summary: Microbiological polyhydroxyalkanoates (PHAs) have great potential as bio-replacements for synthetic polymers. The inherent properties of PHAs expand their applicability in various industries, environments, and clinical sectors. Through omics mining approaches, a new environmental-friendly, endotoxin-free gram-positive bacterium, Bacillus cereus IBA1, was identified as a promising PHA producer. By using a nutrient-enriched strategy, the concentration of PHA granules was increased by 2.3 folds to 2.78 +/- 0.19 g/L. This study also confirmed the growth-dependent PHA biogenesis through exploring PHA granule-associated operons and their constituent genes. The feasibility of this microbial phenomenon could contribute to the development of next-generation biopolymers and increase the industrial applicability of PHAs, thus promoting sustainable development.

Directed Evolution of Sequence-Regulating Polyhydroxyalkanoate Synthase to Synthesize a Medium-Chain-Length-Short-Chain- Length (MCL-SCL) Block Copolymer

Summary: This study successfully improved the activity of the sequence-regulating polyhydroxyalkanoate synthase PhaC(AR) towards a medium-chain-length monomer 3-hydroxyhexanoyl (3HHx)-coenzyme A (CoA) through directed evolution. Beneficial mutations N149D and F314L were identified, which increased the fraction of 3HHx. Furthermore, the F314H mutant enzymatically synthesized a new PHA homopolymer P(3HHx) and enabled the synthesis of a novel PHA block copolymer P(3HHx)-b-P(2HB).

PHA synthase (PhaC) is the key enzyme in PHA biosynthesis. The structure of PhaC consists of an N-terminal domain and a C-terminal catalytic domain, and the N-terminal domain has not been successfully visualized in existing structures.
