RAD-TGTs: high-throughput measurement of cellular mechanotype via rupture and delivery of DNA tension probes

Matthew R. Pawlak et al.

Summary: Mechanical forces drive critical cellular processes, and the authors present Rupture And Deliver Tension Gauge Tethers (RAD-TGTs) which can record the mechanical history of individual cells by using flow cytometry or sequencing. RAD-TGTs recapitulate prior studies on DNA tension probes and can detect fluorescence gain in the force-generating cell. They can differentiate distinct mechanotypes and have the potential for various applications in combination with CRISPR screens and -omics analysis.

Directed cell migration towards softer environments

Aleksi Isomursu et al.

Summary: This study reveals that cells are capable of migrating towards substrates with decreasing stiffness, in addition to the previously known migration towards substrates with increasing stiffness. The migration direction is determined by the optimal stiffness that allows cells to generate maximal traction, rather than the changes in canonical mechanosensitive signaling or actomyosin contractility.

The fibrotic and immune microenvironments as targetable drivers of metastasis

Luke Boulter et al.

Summary: Cancer cells exist within a complex microenvironment of stromal cells and extracellular matrix, where interactions between stromal cell subtypes and signaling pathways play a crucial role in controlling metastasis. The heterogeneity and plasticity within stromal cell populations add to the complexity and provide opportunities for new therapeutic approaches for targeting metastatic disease. Understanding these networks is essential for developing effective treatments.
