BODIPY-based probes for hypoxic environments

Summary: The hypoxic environment is a significant characteristic of solid tumors, resulting from an imbalance between oxygen supply and consumption. Recent progress has been made in designing hypoxia probes derived from BODIPY scaffolds. These probes have attracted particular attention and are closely related to tumor resistance to various therapies. This review focuses on the construction strategies, recognition mechanisms, and biological imaging applications of various BODIPY-based hypoxia probes, aiming to inspire novel approaches and innovations for designing fluorescent probes for imaging hypoxic environments and promoting the development of tumor diagnostics and hypoxia-responsive anti-cancer nanomedicines.

Asymmetric Core-Expanded BOPYOs: Facile Synthesis and Optical Properties

Summary: The study focused on N,O-bidentate BF2 complexes with five- and six-membered core rings, while the development of seven-membered complexes remains unexplored. By utilizing a self-condensation and coordination reaction with BF3·OEt2 as a catalyst, a series of conjugation compounds called BOPYOs were developed from a single starting material. One derivative of BOPYO, which contains a dimethylamino group, exhibited favorable photophysical properties and acid responsiveness based on intramolecular charge transfer effect, making it a potential NIR dye.

N,O-Chelated Organoboron Complexes with Seven-Membered Rings

BODIPY based red emitters: Synthesis, computational and biological studies

Nonsymmetric Benzo[a]fused and Thiophene/Thieno[3,2-b]thiophene[b]fused BODIPYs: Synthesis and Photophysical Properties

The synthesis of nonsymmetric benzo[a]fused and thiophene/thieno[3,2-b]thiophene[b]fused BODIPY derivatives with a reactive site allowed for the development of new compounds through nucleophilic substitution reactions. X-ray crystallographic analysis revealed a planar core structure with pi-pi interactions, and changes in absorption and emission were rationalized through density functional theory calculations.

A computational and experimental investigation of donor-acceptor BODIPY based near-infrared fluorophore for in vivo imaging

Summary: TD-DFT quantum calculation was performed to predict the electronic transition and absorption/emission maxima of pyrrole-difluoroboron derivatives with different electron donor-acceptor units or conjugated degrees. It led to the design of a new NIR fluorophore TPBD-BP, which showed excellent water solubility, biocompatibility, and AIE for in vivo imaging. The NIR-II emission signal of TPBD-BP dots could be observed in the reproductive organs of normal nude mice, indicating potential applications in biological imaging.

Thiophene-Fused BODIPY Dimers and Tetramers from Oxidative Aromatic Couplings as Near-Infrared Dyes

Summary: The study presents a straightforward method to synthesize a family of thiophene-fused BODIPY dimers and tetramers by transforming flexible sulfur bridges into coplanar thiophene fusions. FeCl3 was utilized as a bifunctional oxidant for both intra- and intermolecular oxidative aromatic coupling reactions, leading to strong red-shift absorptions ranging from 509 nm for a BODIPY monomer to 830 nm for a tetramer.
