One-pot synthesis of some new regioselective 4β-pyrazolepodophyllotoxins as DNA topoisomerase-II targeting anticancer agents

Identification of Novel 4′-O-Demethyl-epipodophyllotoxin Derivatives as Antitumor Agents Targeting Topoisomerase II

Summary: C4 variation is an effective strategy to optimize the antitumor spectra of 4'-O-demethyl-epipodophyllotoxin (DMEP). Novel DMEP derivatives with different C4 substituents were synthesized and their antitumor activities were evaluated. Compound 28 displayed strong inhibition against etoposide-resistant KBvin cells and showed promising in vivo antitumor efficacy in a hepatic tumor mouse model. These findings suggest that C4 derivatization of DMEP is a feasible approach to identify potent antitumor agents.

The New Face of a Well-Known Antibiotic: A Review of the Anticancer Activity of Enoxacin and Its Derivatives

Summary: Enoxacin, as a second-generation quinolone antibiotic, exhibits promising anticancer activity through its unique cytotoxic mechanisms including enhanced RNA interference, promotion of microRNA processing, and production of free radicals. It effectively inhibits cancer cell invasiveness by competitively inhibiting vacuolar H+-ATPase subunits and suppressing the c-Jun N-terminal kinase signaling pathway. Newly synthesized derivatives of Enoxacin have shown amplified cytotoxic effects with emphasis on oxidative stress, apoptosis induction, and microRNA interference. Unlike other fluoroquinolones, Enoxacin demonstrates uncommon cytotoxic properties, making it an underestimated anticancer drug with pleiotropic actions.
