pH-Responsive Antimicrobial Peptide with Selective Killing Activity for Bacterial Abscess Therapy

Summary: This study synthesized a series of pH-responsive antimicrobial peptides, among which F5 exhibited outstanding pH-responsive activity and low toxicity. Potentiation of Vancomycin: Creating Cooperative Membrane Lysis through a Derivatization-for-Sensitization Approach

Summary: This study presents a solution to the challenges posed by gram-negative bacteria with multidrug resistance through a novel approach of derivatization-for-sensitization of antibiotics. By modifying vancomycin and using a sensitizing adjuvant, the researchers achieved a significant potentiation of vancomycin and increased effectiveness of the sensitizer. The approach was shown to lyse the bacterial membrane and enhance antibiotic uptake, resulting in improved antimicrobial efficacy in vivo. The potential of this approach was further demonstrated by its application to other antibiotics and sensitizer structures.

Membrane-Active Cyclic Amphiphilic Peptides: Broad-Spectrum Antibacterial Activity Alone and in Combination with Antibiotics

Summary: We designed a library of cyclic peptides containing arginine and tryptophan residues to study their antibacterial activity. Among these peptides, two showed the highest antimicrobial activity and demonstrated synergism when combined with commercially available antibiotics. One of the peptides exhibited selective lethal action against bacteria by disrupting their membranes. It also showed stability in human plasma and minimal potential for the development of resistance in pathogens.

Two plus One: Combination Therapy Tri-systems Involving Two Membrane-Disrupting Antimicrobial Macromolecules and Antibiotics

Summary: This study describes a new combinatorial approach for combating multidrug-resistant bacteria. By combining two different membrane-disrupting antimicrobial agents and an antibiotic, synergistic interactions and biocompatibility were achieved. The combination therapy not only demonstrated bacteriostatic and bactericidal activity but also increased selectivity. The findings shed new insights on the application of membrane-disrupting antimicrobial agents in combination therapy.

Cooperative Membrane Damage as a Mechanism for Pentamidine-Antibiotic Mutual Sensitization

Summary: This study explores the combination therapy of Gram-positive-selective antibiotics and the adjuvant pentamidine against Gram-negative bacteria. Through screening a library of FDA-approved antibiotics, four antibiotics were found to exhibit enhanced activity when combined with pentamidine against multidrug-resistant Gram-negative bacteria. Further research indicates that the physicochemical properties of antibiotics and cooperative membrane damage are key factors for the effectiveness of the combination therapy.

A Dual-Responsive Antibiotic-Loaded Nanoparticle Specifically Binds Pathogens and Overcomes Antimicrobial-Resistant Infections

Summary: A novel nanoparticle coated with dextran and carrying an antibiotic shows strong affinity with various pathogens and effectively accumulates in infected tissues, overcoming antimicrobial resistant infections. The nanoparticle is activated in the infectious microenvironment, releasing polymers and antibiotics that synergistically combat resistant pathogens. This nanoparticle formulation demonstrates safety and efficacy in animal infection models against resistant Gram-negative and Gram-positive pathogens.

Antibiotic Potentiation in Multidrug-Resistant Gram-Negative Pathogenic Bacteria by a Synthetic Peptidomimetic

Summary: The peptidomimetic CEP-136 significantly potentiates the antibacterial activity of clinically important antibiotics against several human pathogens, showing strong synergy with some combinations resulting in over 50-fold reduction in antibiotic concentration. Mechanistic studies suggest the potentiation effect arises from permeabilization of the Gram-negative bacteria's outer membrane lipopolysaccharide layer.
