Summary: Triple-negative breast cancer (TNBC) is an aggressive type accounting for 15%-25% of all breast cancer, which lacks receptors and is unresponsive to hormonal therapies. TNBC patients mainly rely on cytotoxic chemotherapy. Activation of Wnt pathways is associated with mammary hyperplasia and tumor formation. Tumor suppressor microRNAs can inhibit tumor cell growth, invasion, and migration, as well as down-regulate Wnt signaling. Nanoparticles containing microRNA have shown enhanced effectiveness compared to single release. This review explores the role of Wnt signaling in TNBC, EMT, metastasis, anti-drug resistance, and microRNA regulation of Wnt, as well as the use of nano-carriers for micro-RNA delivery and clinical biomarkers related to Wnt pathways.

Summary: Marine macroalgae or seaweeds, specifically Codium tomentosum and Actinotrichia fragilis from the Red Sea, were investigated for their antibacterial and antioxidant activities. The results showed that Codium tomentosum extract exhibited superior antibacterial and antioxidant effects compared to Actinotrichia fragilis. Additionally, fillet fish fortified with Codium tomentosum extract had the highest overall acceptance score, indicating its potential as a food supplement. These findings suggest that these seaweeds can be cultivated as a sustainable and safe source of therapeutic compounds for treating human and fish diseases.

Brefeldin A from the Deep-Sea-Derived Fungus Fusarium sp. Targets on RIPK3 to Inhibit TNFα-Induced Necroptosis

Summary: In this study, four known compounds were isolated from deep-sea-derived Fusarium sp. ZEN-48, and their structures were determined. Among them, it was found that one of the compounds, BFA, showed significant inhibition on TNF alpha-induced necroptosis by disrupting necrosome formation and suppressing RIPK3 and MLKL phosphorylation. This finding has important implications for the treatment of necroptosis-related inflammatory diseases and the development of new drugs.

New brefeldin A-cinnamic acid ester derivatives as potential antitumor agents: Design, synthesis and biological evaluation

Summary: This study designed and synthesized a series of BFA-cinnamic acid ester derivatives, and evaluated their in vitro cytotoxic activities on different cancer cell lines. The most potent derivative 33 showed improved aqueous solubility and strong cytotoxic activity against HepG2 and BEL-7402 cell lines, while exhibiting low toxicity on normal liver cell line L02. Mechanistic studies revealed that 33 achieved its anti-tumor effects by blocking the cell cycle and inducing apoptosis, and displayed favorable pharmacokinetic properties.

Brefeldin A delivery nanomicelles in hepatocellular carcinoma therapy: Characterization, cytotoxic evaluation in vitro, and antitumor efficiency in vivo

Summary: This study encapsulated the macrolide brefeldin A (BFA) in mixed nanomicelles to improve its solubility, stability, and sustained release capability. In a HepG2 tumor-bearing xenograft mouse model, M-BFA effectively delayed tumor progression and induced extensive necrosis.
