Astaxanthin attenuates cigarette smoking-induced oxidative stress and inflammation in a sirtuin 1-dependent manner

Mingming Deng et al.

Summary: Astaxanthin (AXT) can alleviate pulmonary emphysema and small airway fibrosis in chronic obstructive pulmonary disease (COPD) by regulating MMP-9/TIMP-1 expression, inhibiting oxidative stress, and the inflammatory pathway. AXT can directly bind to SIRT1, regulate its deacetylation activity, and reduce oxidative stress by activating antioxidant enzymes and suppressing the inflammatory response.

Cellular and Molecular Signatures of Oxidative Stress in Bronchial Epithelial Cell Models Injured by Cigarette Smoke Extract

Chiara Cipollina et al.

Summary: Exposure of airway epithelium to environmental insults, such as cigarette smoke, leads to increased oxidative stress, which promotes the development of chronic lung diseases. Oxidative stress disrupts antioxidant defenses, affects cellular autophagy and survival regulatory mechanisms, leading to cell senescence. These events are exacerbated by inflammation driven by oxidative stress. Various models of bronchial epithelial cells are used to study the mechanisms and functions altered by cigarette smoke exposure and to test the efficacy of antioxidants. This review provides a comprehensive synthesis of in-vitro and ex-vivo studies from 2011 to 2021, investigating the molecular and cellular mechanisms triggered by cigarette smoke exposure in bronchial epithelial cells, commonly used experimental models, and the mechanisms of action of cellular antioxidant systems, as well as natural and synthetic antioxidant compounds.
