Synergistic ROS generation and directional overloading of endogenous calcium induce mitochondrial dysfunction in living cells

Mitochondrial Ca2+-overloading by oxygen/glutathione depletion-boosted photodynamic therapy based on a CaCO3 nanoplatform for tumor synergistic therapy

Summary: A CaCO3-based nano-platform responsive to the tumor microenvironment was designed. The released CAT and BSO relieved tumor hypoxia, enhanced PDT efficacy, and induced cell apoptosis, while also destroying the Ca2+ buffering capacity leading to mitochondrial Ca2+ overloading. The synergistic effect of enhanced PDT and mitochondrial Ca2+ overloading showed remarkable antitumor performance.

Double-activation of mitochondrial permeability transition pore opening via calcium overload and reactive oxygen species for cancer therapy

Summary: In this study, a mitochondrial pathway was established to inhibit tumor growth through the double activation of the mitochondrial permeability transition pore (MPTP) channel. Fe2+ doped covalent organic frameworks (COF) were synthesized and used as templates to grow a CaCO3 shell, resulting in the formation of O-2-FeCOF@CaCO3 nanocomposite. After modification with folic acid (FA), the nanocomposite (OFCCF) could target breast cancer cells and achieve synergistic treatment through photodynamic therapy (PDT) and calcium overload. The multifunctional nanosystem with tumor microenvironment (TME) response abilities provided a novel strategy for clinical cancer therapy.

Engineering of a bona fide light-operated calcium channel

Summary: The current optogenetic toolkit lacks a robust single-component Ca2+-selective ion channel tailored for remote control of Ca2+ signaling in mammals. Existing tools are either derived from engineered channelrhodopsin variants without strict Ca2+ selectivity or based on the stromal interaction molecule 1 (STIM1) that might crosstalk with other targets. Here, we describe the design of a light-operated Ca2+ channel (designated LOCa) by inserting a plant-derived photosensory module into the intracellular loop of an engineered ORAI1 channel. LOCa displays biophysical features reminiscent of the ORAI1 channel, which enables precise optical control over Ca2+ signals and hallmark Ca2+-dependent physiological responses. Furthermore, we demonstrate the use of LOCa to modulate aberrant hematopoietic stem cell self-renewal, transcriptional programming, cell suicide, as well as neurodegeneration in a Drosophila model of amyloidosis. Existing optogenetic methods to induce calcium mobilisation lack selectivity and specificity. Here, the authors design and engineer a single-component light-operated calcium channel to provide optical control over calcium signals and calcium-dependent physiological responses: LOCa.

Upconverted Metal-Organic Framework Janus Architecture for Near-Infrared and Ultrasound Co-Enhanced High Performance Tumor Therapy

Summary: A biocatalytic Janus nanocomposite, UPFB, was fabricated for ultrasound-driven SDT and 808 nm NIR light-mediated PDT by combining UCNPs and a ferric zirconium porphyrin MOF. UPFB not only promotes ROS generation via GSH depletion and oxygen supply from Fe3+ ions but also achieves CDT performance under acidic conditions. Additionally, UPFB linked with biotin exhibits a good targeting ability to rapidly accumulate in the tumor region, making it suitable for cancer therapeutics.

A Robust ROS Generation Strategy for Enhanced Chemodynamic/Photodynamic Therapy via H2O2/O2 Self-Supply and Ca2+ Overloading

Summary: The smart polyethylene glycol (PEG)-ylated nanosystem CaO2@ZIF-Fe/Ce6@PEG (CaZFCP) is designed to supply H2O2/O-2 and cause Ca2+ overloading in tumor cells, enhancing the efficacy of chemodynamic therapy (CDT) and photodynamic therapy (PDT) simultaneously. The CaZFCP releases CaO2 nanoparticles, Fe2+, and photosensitizer Chlorin e6 (Ce6) in the weakly acidic tumor microenvironment, alleviating the deficiency of endogenous H2O2 and hypoxia in the tumor area, leading to enhanced production of hydroxyl radicals and singlet oxygen for improved CDT and PDT. Additionally, the Ca2+ overloading induced by CaO2 decomposition amplifies intracellular oxidative stress, resulting in mitochondrial dysfunction and further enhancing the efficacy of combined CDT/PDT. The in vitro and in vivo experimental results confirm the excellent tumor inhibition effect of CaZFCP, providing a simple paradigm for ROS-involved cancer therapies.
