Isolation, characterisation, anticancer and anti-oxidant activities of 2-methoxy mucic acid from Rhizophora apiculata: an in vitro and in silico studies

Summary: This study isolated and characterized a novel bioactive molecule, 2-methoxy mucic acid, from the leaves of Rhizophora apiculate Blume. The molecule exhibited promising anticancer activity against HeLa and MDA-MB-231 cancer cell lines, as well as antioxidant activity. In addition, it showed high binding affinity towards the Bcl-B protein and improved its structural stability. This research provides important insights into the significance of 2-methoxy mucic acid and opens up possibilities for the development of anticancer drugs.

Analysis of circRNA profiles and clinical value in Stevens-Johnson syndrome and toxic epidermal necrolysis

Summary: This study analyzed the expression profiles of circRNAs in peripheral blood mononuclear cells of SJS/TEN patients and found a correlation between circRNA expression and disease severity. Specifically, hsa_circ_0083619 was identified as a potential indicator for SJS/TEN severity and a circRNA-miRNA-mRNA network was constructed. The study suggests that the hsa_circ_0083619/miR-18a-5p/BCL2L10 axis may play a role in the pathogenesis of SJS/TEN.

Summary: Overexpression of Bcl-2 proteins like Bcl2L10 is linked to chemotherapy resistance and poor survival in several types of cancer. A particular SNP (rs2231292) in the BCL2L10 gene has been found to lower resistance to chemotherapy, leading to improved survival in acute leukemia and colorectal cancer patients. This study investigated the impact of this SNP in breast cancer, finding that individuals with the Nrh-R isoform of Bcl2L10 had higher sensitivity to cell death induction and potentially better outcomes.

Targeting MCL-1 protein to treat cancer: opportunities and challenges

Summary: Evading apoptosis through overexpression of prosurvival BCL-2 family proteins, such as Mcl-1, is a mechanism that contributes to tumor development and chemoresistance. Targeting MCL-1 protein has shown success in inducing apoptosis and overcoming resistance to chemotherapy and targeted therapy. Various strategies to inhibit MCL-1's antiapoptotic activity have been explored, including targeting its interactions with other proteins.

Expression patterns and biological function of BCL2L10 during mouse preimplantation development

Summary: This study investigated the expression and function of Bcl2l10 during mouse preimplantation development. The results showed that Bcl2l10 was highly expressed in metaphase II-stage oocytes and pronuclear-stage embryos. Knocking down Bcl2l10 resulted in reduced blastocyst formation rate and decreased expression of OCT4, NANOG, and SOX17.

Last but not least: BFL-1 as an emerging target for anti-cancer therapies

Summary: BFL-1, an understudied pro-survival BCL-2 protein, is expressed in many cancers. However, it is unclear whether high transcript expression always correlates with cell survival function. Recent research suggests that BFL-1 may be a resistance factor in blood cancers, highlighting the importance of understanding its role and resistance mechanisms in human cancers. Currently, there are no effective BFL-1 inhibitors available.

It acts as a pro-survival factor by protecting cells from the cytotoxic effects of various drugs and inhibitors, suggesting that targeting BCL2L10 may enhance the clinical efficacy of other therapies for malignant melanoma.

Summary: BH3 mimetics are a new class of anticancer drugs that target prosurvival BCL2 proteins to induce apoptosis, with Venetoclax being the first approved BCL2 inhibitor showing activity in various lymphoid and myeloid neoplasms. Long-term exposure may lead to secondary resistance. Success of BCL2 inhibitors has led to development of BH3 mimetics targeting MCL1, with promising preclinical activity but potential challenges due to MCL1's physiological roles in nonhematologic tissues.
