The Devastating Clinical Consequences of Child Abuse and Neglect: Increased Disease Vulnerability and Poor Treatment Response in Mood Disorders

Elizabeth T. C. Lippard et al.

Summary: A large body of evidence has demonstrated the long-term consequences of childhood maltreatment, which increases the risk for psychiatric and medical disorders. This review summarizes the effects of childhood maltreatment on vulnerability to mood disorders, highlighting the increased prevalence and associated risks. It discusses the persistent alterations in the hypothalamic-pituitary-adrenal axis and inflammatory cytokines, as well as candidate genes, environmental factors, and neurobiological associations that may mediate these effects. The studies provide insights into modifiable mechanisms and guide treatment and prevention strategies.

Corticolimbic DCC gene co-expression networks as predictors of impulsivity in children

Jose M. Restrepo-Lozano et al.

Summary: Inhibitory control deficits are common in neuropsychiatric conditions, and early markers of vulnerability to this behavioral trait are yet to be discovered. The maturation of the prefrontal cortex and the development of inhibitory control are closely related. The DCC gene is involved in corticolimbic development and is associated with impulsive phenotypes in children, as shown by the ePRS score.

Mapping genomic loci implicates genes and synaptic biology in schizophrenia

Vassily Trubetskoy et al.

Summary: In this study, a two-stage genome-wide association study was conducted to identify common variants associated with schizophrenia. The results revealed 287 distinct genomic loci and 120 genes likely to be involved in the development of schizophrenia. This research provides valuable insights into the pathophysiology of schizophrenia and offers a resource for further mechanistic studies.

Dissecting diagnostic heterogeneity in depression by integrating neuroimaging and genetics

Amanda M. Buch et al.

Summary: Depression is a complex psychiatric syndrome with multiple pathophysiological mechanisms. Large-scale genome-wide association studies have identified genetic risk variants implicating excitatory neurotransmission and synapse function, highlighting a highly polygenic inheritance pattern as an important contributor to the heterogeneity in depression.
