1,25-Dihydroxyvitamin D Deficiency Accelerates Aging-related Osteoarthritis via Downregulation of Sirt1 in Mice

Summary: Emerging observational data suggest that vitamin D deficiency is associated with knee osteoarthritis. However, the relationship between vitamin D level and osteoarthritis, as well as the role of vitamin D supplementation in prevention, are still controversial. This study found that 1,25(OH)2D3 deficiency accelerated the development of knee osteoarthritis in mice, while supplementation with 1,25(OH)2D3 rescued the phenotypes of osteoarthritis.

Dysfunction of metabolic activity of bone marrow mesenchymal stem cells in aged mice

Summary: This study investigated the metabolic activity of MSCs and its possible mechanism during aging. The findings suggest that the metabolic function of BMSCs is impaired in aged mice, characterized by decreased oxidative phosphorylation and glycolytic activity, as well as lower mitochondrial membrane potential, NAD+/NADH ratio, and ATP production. FABP4 may play a key role in BMSC senescence caused by fatty acid metabolism disorders. Regulation of metabolic activity could be a potential therapeutic target for enhancing the regenerative functions of BMSCs.

Interleukin-6 Knockout Inhibits Senescence of Bone Mesenchymal Stem Cells in High-Fat Diet-Induced Bone Loss

Summary: IL-6 KO mice exhibited reduced weight gain and inhibited bone loss, expansion of bone marrow adipose tissue, adipogenesis in bone marrow, and bone formation under high-fat diet conditions. IL-6 KO also suppressed the clone formation of bone marrow cells and the expression of senescence markers. Additionally, IL-6 antibody inhibited the activation of STAT3 and senescence in bone mesenchymal stem cells, while rescuing IL-6 induced senescence in IL-6 KO mice through the STAT3/p53/p21 pathway.
