Selective xyloglucan oligosaccharide hydrolysis by a GH31 α-xylosidase from Escherichia coli

Summary: In this study, we investigated the hydrolysis of xyloglucan oligosaccharides (XGOs) by the Escherichia coli alpha-xylosidase (YicI). Our results showed that YicI preferentially removes the xylosyl residue from non-galactosylated oligosaccharides. Molecular dynamics simulations and site-directed mutagenesis experiments revealed that hydrogen bond interactions between D49 and galactosylated oligosaccharides are important for enzyme-XGO interactions.
