Expansion of macrophage and liver sinusoidal endothelial cell subpopulations during non-alcoholic steatohepatitis progression

Zhenyang Shen et al.

Summary: Liver non-parenchymal cells (NPCs) are important for the progression of non-alcoholic steatohepatitis (NASH). Through single-cell RNA sequencing, we found proinflammatory subpopulations of macrophages and liver sinusoidal endothelial cells (LSECs) that may contribute to NASH development. The expansion of these subpopulations was associated with NASH severity, suggesting their involvement in NASH progression.

MyD88 in hepatic stellate cells enhances liver fibrosis via promoting macrophage M1 polarization

Jie Zhang et al.

Summary: During liver fibrosis, the MyD88 signaling in HSCs plays a crucial role by promoting the activation of HSCs and contributing to the development of liver fibrosis. Inhibition of MyD88 signaling can attenuate liver fibrosis, reduce inflammatory response, and provide a promising therapeutic target for the treatment of liver fibrosis.

Reactive Oxygen Species and Oxidative Stress in the Pathogenesis of MAFLD

Kathleen Clare et al.

Summary: The pathogenesis of MAFLD is complex and involves multiple parallel hits on a background of genetic susceptibility. Excessive hepatic lipid accumulation leads to mitochondrial dysfunction, endoplasmic reticular stress, and ROS overproduction. The use of antioxidants and ROS scavengers can alleviate the proapoptotic effects of fatty acids. Activation of Nrf2, THR beta agonists, and nuclear PPAR family can reduce oxidative stress and inflammation in MAFLD models. Multiomics-based strategies, including transcriptome profiling and single-cell transcriptome analysis, provide hope for identifying novel insights and therapeutic targets in MAFLD progression.
