Enhanced surface nanoanalytics of transient biomolecular processes

Summary: In this study, N-terminal E cyclization was identified as the major degradation pathway for monoclonal antibodies and bispecific antibodies in lyophilized formulations. The use of salt-gradient ion exchange chromatography and hydrophobic interaction chromatography effectively separated pE, and pE did not affect the binding activity of the antibodies. In vitro experiments suggested a potential role of serum matrix in pE conversion.

Studies showed the presence of pGlu79-alpha-synuclein in brain samples of PD and dementia with Lewy body subjects, with a spatial co-occurrence with the enzyme QC.

Examples like PFVYLI, a hydrophobic CPP derived from alpha 1-antitrypsin, show self-assembling properties and noncovalent complex formation with nucleic acids. Despite lacking cationic residues in the composition, PFVYLI forms complexes that retain amyloid features and promote nucleic acid internalization, suggesting potential applications in nanostructured scaffolds.
