Activity of Fatty Acid Biosynthesis Initiating Ketosynthase FabH with Acetyl/Malonyl-oxa/aza(dethia)CoAs

Trevor J. Boram et al.

Summary: Fatty acid and polyketide biosynthetic enzymes use acyl- and malonyl-thioesters for catalysis. This study focuses on FabH, which initiates fatty acid biosynthesis in bacteria and plants. By synthesizing analogues of acetyl/ malonyl-CoA, this study evaluates their stability and behavior as substrates or inhibitors for FabH, providing insight into the enzyme/substrate interactions.

Conformational Selection Governs Carrier Domain Positioning in Staphylococcus aureus Pyruvate Carboxylase

Joshua H. Hakala et al.

Summary: The study developed a system to directly observe the positioning of the carrier domain in PC in the presence or absence of ligands, independent of catalytic turnover. It demonstrated that the equilibrium of carrier domain positioning is sensitive to substrate analogues and the allosteric activator, controlled by conformational selection.

CryoEM structural exploration of catalytically active enzyme pyruvate carboxylase

Jorge Pedro Lopez-Alonso et al.

Summary: This study investigates the structure of active pyruvate carboxylase tetramers using cryoEM and reveals the catalytic steps and substrate-product identification. The analysis of catalytically active PC tetramers uncovers the role of certain motions during enzyme functioning and the mechanism for allosteric regulation.

The role of the phosphate groups of trinitrophenyl adenosine 5′-triphosphate (TNP-ATP) in allosteric activation of pyruvate carboxylase and the inhibition of acetyl CoA-dependent activation

Khanti Rattanapornsompong et al.

Summary: This study investigated the effects of the triphosphate group of TNP-ATP and its derivatives on the allosteric activation of the enzyme and inhibition of acetyl-CoA-dependent activation. The findings suggest that reducing the number of phosphate groups in TNP-ATP derivatives decreases their activation efficacy and affects the binding affinity and inhibition of enzyme activation by acetyl-CoA. These results provide insight into potential drug development for metabolic diseases and cancers with abnormal expression of key enzymes.
