Selective motor activation in organelle transport along axons

Sydney E. Cason et al.

Summary: The active transport of organelles and other cargos along the axon is essential for maintaining neuronal health and function. However, the complex regulatory mechanisms involved in this process are still not fully understood. This review discusses the current understanding of motor regulation in axonal transport and explores how regulatory proteins affect the activity of dynein or kinesin motors. The specific mechanisms that regulate motor activity during organelle trafficking in the axon are also highlighted.

Mechanochemical tuning of a kinesin motor essential for malaria parasite transmission

Tianyang Liu et al.

Summary: In this study, the authors investigated the role of Plasmodium kinesin-8B in microtubule motility and depolymerization. They found that kinesin-8B is essential for the formation of male gametes and parasite transmission. The study provides important insights into the molecular mechanisms underlying malaria transmission.

Kinesin-8-specific loop-2 controls the dual activities of the motor domain according to tubulin protofilament shape

Byron Hunter et al.

Summary: This study reveals how Kinesin-8 alternates between promotility and pro-microtubule-depolymerization states by regulating its tubulin structure through the loop-2 region. Structural changes induced by curved microtubule ends and straight protofilaments result in different functional outcomes.

Kinesin-binding protein remodels the kinesin motor to prevent microtubule binding

April L. Solon et al.

Summary: KIFBP is found to remodel kinesin motors and block microtubule binding by inducing allosteric changes to kinesin and sterically blocking access to the microtubule. The study also identifies two regions of KIFBP essential for kinesin binding and cellular regulation during mitosis.

Structural basis of mechano-chemical coupling by the mitotic kinesin KIF14

Matthieu P. M. H. Benoit et al.

Summary: The study presents the structural changes of KIF14 bound to microtubules, revealing the crucial roles of microtubule binding, nucleotide species, and the neck-linker domain in the transition between major conformations of the motor domain. Additionally, the position of the neck-linker regulates hydrolysis rather than nucleotide binding step.
