Exosome-derived circCCAR1 promotes CD8+T-cell dysfunction and anti-PD1 resistance in hepatocellular carcinoma

Zongqiang Hu et al.

Summary: The study found that circCCAR1, a circular RNA, is present at high levels in tumor tissues and plasma of hepatocellular carcinoma (HCC) patients. CircCCAR1 promotes the growth and metastasis of HCC through the miR-127-5p/WTAP signaling pathway, and causes CD8+ T-cell dysfunction. Moreover, circCCAR1 enhances resistance to anti-PD1 immunotherapy by stabilizing the PD-1 protein and impairing CD8+ T-cell function.

LncRNA LINC00184 promotes docetaxel resistance and immune escape via miR-105-5p/PD-L1 axis in prostate cancer

Wei Zhang et al.

Summary: The study revealed that LINC00184 is associated with docetaxel resistance and immune escape in prostate cancer cells. By adsorbing miR-105-5p, LINC00184 promotes these phenomena and subsequently inhibits the expression of PD-L1. This indicates that LINC00184 may serve as a potential target for treatment in prostate cancer patients with docetaxel resistance.

Summary: Early clinical data suggest that some patients with castration-resistant prostate cancer may benefit from PD-L1 inhibition, especially with enzalutamide. However, a trial combining atezolizumab with enzalutamide did not improve overall survival in unselected patients. Genomic and immune biomarkers associated with response to immune checkpoint inhibitors were identified.

Docetaxel resistance-derived LINC01085 contributes to the immunotherapy of hormone-independent prostate cancer by activating the STING/MAVS signaling pathway

Summary: This study found that LINC01085, as a tumor suppressor, has significant clinical relevance in patients with aggressive hormone-independent prostate cancer (HIPC) who have acquired docetaxel resistance. The overexpression of LINC01085 inhibits TBK1 dimerization and affects the interaction between TBK1 and GSK313, resulting in changes in the expression levels of PD-L1, NF-xB, and interferons. Overexpression of LINC01085 combined with immune checkpoint blockade is proposed as an effective strategy for treating HIPC patients.

High dose androgen suppresses natural killer cytotoxicity of castration-resistant prostate cancer cells via altering AR/circFKBP5/miRNA-513a-5p/PD-L1 signals

Min Tang et al.

Summary: In castration-resistant prostate cancer (CRPC), high dose dihydrotestosterone (DHT) suppresses the ability of natural killer (NK) cells to inhibit CRPC cells and negatively impacts immune surveillance. Mechanistically, DHT transactivates AR to increase circularRNA-FKBP5 (circFKBP5) expression, which inhibits miR-513a-5p and increases PD-L1 expression. Targeting PD-L1 enhances the suppressive effect of high dose DHT on CRPC cell growth.

DC-based immunotherapy, such as Sipuleucel-T, has shown improved overall survival in prostate cancer, but further research into DC vaccines has been limited. Understanding the immunosuppressive environment of prostate cancer and developing new vaccine strategies that can overcome this environment is essential for future success in immunotherapy.
