An atlas of substrate specificities for the human serine/threonine kinome

Summary: Protein phosphorylation is a common post-translational modification, and thousands of phosphorylated sites have been associated with human diseases and biological processes. However, the responsible protein kinases for most phosphorylation events are currently unknown. This study used synthetic peptide libraries to profile the substrate specificity of 303 Ser/Thr kinases and identified the kinases capable of phosphorylating reported phosphorylation sites.

Summary: Diabetes, insulin resistance, and obesity are risk factors for Alzheimer's disease. In this study, the PDK-1 agonist PS48 was orally administered to transgenic mice and showed positive effects on learning and memory. PS48 was particularly effective in improving spatial memory in mice on a high fat diet.

RSK inhibitors as potential anticancer agents: Discovery, optimization, and challenges

Summary: Ribosomal S6 kinase (RSK) is a group of serine/threonine kinases that play an important role in cell growth, proliferation, and migration, as well as tumor occurrence and development. Although several RSK inhibitors have been discovered or designed, only two have entered clinical trials, with limitations of low specificity, low selectivity, and poor pharmacokinetic properties. Further design focuses on selectivity due to functional differences among RSK isoforms. This review summarizes the types of cancers associated with RSK, structural characteristics and optimization process of reported RSK inhibitors, and discusses the importance of selectivity and future drug development directions.

Summary: This study investigates the association-dissociation kinetics of the PDK1:PKCa heterodimer. The findings reveal that single PDK1 molecules freely diffusing on the membrane surface undergo reversible switching between the PDK1 monomer and the PDK1:PKCa heterodimer. The elucidation of these kinetics has important biological and medical implications.
