Autolysis-induced extracellular production of intracellular carboxylesterase EstGtA2 using multiple-protease-deficient Bacillus subtilis strains

Efficient monoacylglycerol synthesis by carboxylesterase EstGtA2 from Geobacillus thermodenitrificans in a solvent-free two-phase system

Summary: This study investigated high-yield monoacylglycerol (MAG) synthesis in a solvent-free two-phase system using bacterial lipolytic enzymes. The study found that a high glycerol/fatty acid molar ratio, the carboxylesterase EstGtA2, and neutralizing the pH of the reaction solution were important strategies for achieving high-yield MAG synthesis. Furthermore, a thermal-tolerant variant of EstGtA2 (EstGtA2(S26I)) showed improved stability and increased the MAG synthesis rate. The addition of alkali and the use of a thermal-tolerant enzyme were crucial for obtaining high-yield MAG.

Summary: In this study, we successfully secreted the Lysobacter enzymogenes beta-lytic protease (BLP) in Bacillus subtilis, marking the first report of heterologous production of BLP in its active form, providing potential for industrial applications. Additionally, a new strategy of utilizing the extracellular proteases of B. subtilis for maturation of heterologous proteins was proposed in this study.
