Development of a three-dimensional blood-brain barrier network with opening capillary structures for drug transport screening assays

Summary: The blood-brain barrier (BBB) is a selective barrier that regulates the transport of solutes in the central nervous system. Current in vitro models cannot replicate the complexity of the brain microvasculature. In this study, a 3D self-assembled microvascular network model was developed, which demonstrated similar permeability to the actual in vivo brain. The model also confirmed the activity of P-glycoprotein efflux pump and the functionality of receptor-mediated transcytosis. The BBB model with capillary openings could be a valuable tool for drug screening.
