Unsaturated bond recognition leads to biased signal in a fatty acid receptor

Summary: In this study, six cryo-electron microscopy structures of GPR120 in complex with FA hormones or TUG891 and Gi or Giq trimers were reported. The study also investigated synthetic ligand selectivity and the structural basis of missense single-nucleotide polymorphisms. Tethered peptide activation mechanism of the adhesion GPCRs ADGRG2 and ADGRG4

Summary: In this study, three cryo-electron microscopy structures of aGPCRs coupled to the G(s) heterotrimer are presented, revealing the structural and biochemical insights into the tethered activation mechanism of aGPCRs. The structures of ADGRG2-beta-G(s) and ADGRG4-beta-G(s) complexes, as well as the ADGRG2(FL)-IP15-G(s) complex, provide valuable information about the binding and activation of aGPCRs by the Stachel sequences. Additionally, rational design of peptidic agonists and generation of peptidic antagonists for aGPCRs based on the aGPCR-beta structures are demonstrated.

Structural basis for the tethered peptide activation of adhesion GPCRs

Summary: This study reports the cryo-EM structures of two aGPCRs in complex with G(s): GPR133 and GPR114. The structures reveal that the Stachel sequences of both receptors assume an alpha-helical-bulge-beta-sheet structure and insert into a binding site formed by the transmembrane domain (TMD). The study also provides insights into the mechanisms of Stachel sequence-mediated receptor activation and Gs coupling.
