PLIN5 interacts with FATP4 at membrane contact sites to promote lipid droplet-to-mitochondria fatty acid transport

Gregory E. Miner et al.

Summary: Cells regulate their metabolism by remodeling membrane contact sites that direct metabolites to different fates. Lipid droplet (LD)-mitochondria contacts change in response to fasting, cold exposure, and exercise. This study focuses on PLIN5, an LD protein that attaches mitochondria, to investigate the function and regulation of LD-mitochondria contacts. The findings reveal that phosphorylation of PLIN5 promotes efficient fatty acid trafficking from LDs to mitochondria during starvation and requires the intact mitochondrial tethering domain of PLIN5. The study also identifies FATP4 as a mitochondrial interactor of PLIN5 and demonstrates that the interaction between the C-terminal domains of PLIN5 and FATP4 induces organelle contacts.

CD36 promotes de novo lipogenesis in hepatocytes through INSIG2-dependent SREBP1 processing

Han Zeng et al.

Summary: This study found that CD36 plays a role in the development of NAFLD by facilitating the uptake of free fatty acids and enhancing de novo lipogenesis in hepatocytes. CD36 forms a complex with INSIG2, disrupting the interaction between SCAP and INSIG2, leading to the translocation of SREBP1 for processing. These findings provide new evidence for targeting CD36 as a potential therapeutic approach for NAFLD.

FATP4 inactivation in cultured macrophages attenuates M1-and ER stress-induced cytokine release via a metabolic shift towards triacylglycerides

Yuling Zhang et al.

Summary: FATP4 deficiency shifts macrophage lipid metabolism towards triacylglyceride synthesis, protecting cells from inflammatory cytokine release and cellular injury. Additionally, lack of FATP4/Fatp4 reduces ceramide levels, induces CD206 up-regulation, and attenuates IL-8, TNF-alpha, and IL-12 release.

Polymorphisms rs55710213 and rs56334587 regulate SCD1 expression by modulating HNF4A binding

Summary: The study showed that SCD1 is directly regulated by HNF4A through binding to key regulatory regions in the SCD1 gene. Knocking down HNF4A significantly decreased SCD1 expression, indicating a crucial role of HNF4A in regulating SCD1 activity and metabolic pathways associated with diseases.
