Liver tryptophan 2,3-dioxygenase: a determinant of anxiety-like behaviour - studies with chronic nicotine administration in rats

Summary: Serotonin deficiency in major depressive disorder (MDD) has long been considered the basis for antidepressant drug development. However, recent evidence challenges the role of inflammation in serotonin deficiency, and suggests that inhibition of liver TDO and KMO should be the focus of MDD pharmacotherapy. Antidepressants that have anti-inflammatory properties may be effective in blocking the activation of KMO.

Inflammation and serotonin deficiency in major depressive disorder: molecular docking of antidepressant and anti-inflammatory drugs to tryptophan and indoleamine 2,3-dioxygenases

Summary: The present study considers the roles of the kynurenine pathway (KP) of tryptophan degradation in major depressive disorder (MDD) and the associated inflammatory state. Antidepressants were found to bind to tryptophan 2,3-dioxygenase (TDO) but not to indoleamine 2,3-dioxygenase (IDO). TDO inhibition is a common property of antidepressants, while IDO induction may play a role in defective serotonin synthesis in MDD. Antidepressants with anti-inflammatory properties may inhibit IDO induction by lowering proinflammatory cytokine levels. The interaction between serotonergic and glutamatergic activities may underpin the pathophysiology of MDD.
