Summary: Antineoplastic drug oxaliplatin induces memory and emotional deficits, accompanied by synaptic dysfunction and downregulation of NR2B subunit in the hippocampus. Chronic oral administration of magnesium-L-threonate (L-TAMS) prevents these behavioral and synaptic changes, which are associated with reduced brain magnesium levels. These findings suggest that TNF-alpha/NF-kappa B signaling activation due to magnesium deficiency is responsible for the memory and emotional deficits induced by oxaliplatin, and L-TAMS may be a novel approach to treating chemotherapy-induced cognitive impairments.

Factors such as physical exercise and environmental stimulation may protect against chemotherapy-induced neurogenic suppression and hippocampal neurotoxicity. Pharmacological interventions targeting the hippocampus are being reviewed to prevent or reduce the cognitive and neurotoxic side effects of chemotherapy.
