Increased mitophagy protects cochlear hair cells from aminoglycoside-induced damage

Summary: This study found that aminoglycosides cause hair cell injury by impairing mitophagy, a process that degrades damaged mitochondria. The impaired mitophagy is characterized by blocked recruitment of PRKN to mitochondria and phagophore recognition of damaged mitochondria. Additionally, neomycin disrupts mitophagy by inhibiting Pink1 expression and inducing ATF3 expression. Treatment with a mitophagy activator can rescue neomycin-treated hair cells by increasing mitophagy. Overall, this study suggests that modulating or intervening in mitophagy may have therapeutic potential for aminoglycoside-induced hearing loss.

DJ-1 Protects auditory cells from cisplatin-induced ototoxicity via regulating apoptosis and autophagy

Summary: This study aimed to investigate the expression of DJ-1 in auditory cells and its correlation with cisplatin-induced ototoxicity. DJ-1 expression was detected in mouse cochlea hair cells and HEI-OC1 cells, and was reduced by cisplatin treatment. DJ-1 participated in cisplatin-induced cell damage by regulating apoptosis and autophagy. These findings provide a new strategy for the prevention of cisplatin-induced ototoxicity.

c-Myb protects cochlear hair cells from cisplatin-induced damage via the PI3K/Akt signaling pathway

Summary: The expression of c-Myb is downregulated in cochlear cells after cisplatin damage, and overexpression of c-Myb can increase cell viability, reduce apoptosis, and decrease reactive oxygen species levels. Knockout of c-Myb exacerbates cisplatin-induced cochlear cell loss, apoptosis, and hearing deficits. Mechanistic studies reveal that c-Myb contributes to increased cell survival after cisplatin exposure by upregulating the PI3K/Akt signaling pathway.

Kanamycin and Cisplatin Ototoxicity: Differences in Patterns of Oxidative Stress, Antioxidant Enzyme Expression and Hair Cell Loss in the Cochlea

Summary: Kanamycin and cisplatin are ototoxic drugs with unclear mechanisms of cochlear damage. Kanamycin causes auditory threshold shifts and outer hair cell loss, as well as apoptosis and oxidative stress. In contrast, cisplatin induces different injuries in different regions of the cochlea, with more outer hair cells surviving in the apical region compared to kanamycin, despite the absence of evoked auditory activity. Additionally, there are differential regulations of antioxidant enzyme levels between the two drugs.

Global, regional, and national burden of diseases and injuries for adults 70 years and older: systematic analysis for the Global Burden of Disease 2019 Study

Summary: Based on the data analysis of GBD 2019, the global population of older adults has increased since 1990, and overall mortality rates have decreased for both men and women. However, mortality rates due to falls have increased during the same period. The probability of death among people aged 70-90 has decreased, mainly due to reductions in non-communicable diseases. Globally, disability burden is mainly driven by functional decline, vision and hearing loss, and symptoms of pain. Life expectancy at age 70 (LE-70) and healthy life expectancy at age 70 (HALE-70) have shown continuous increases since 1990, with regional disparities. Higher LE-70 results in higher HALE-70 and slightly increased proportion of years in ill health at age 70 (PYIH-70). Sociodemographic factors, healthcare access and quality indices are positively correlated with HALE-70 and LE-70. Limited data are available for various dietary, environmental or occupational, and metabolic risks.

The antioxidant enzyme PRDX1 plays a crucial role in modulating autophagy in SGNs, which in turn activates the PTEN-AKT signaling pathway to trigger autophagy. These findings suggest potential therapeutic targets for drug-induced SNHL through autophagy activation.
