Anemonin reduces hydrogen peroxide-induced oxidative stress, inflammation and extracellular matrix degradation in nucleus pulposus cells by regulating NOX4/NF-κB signaling pathway

Zhijia Ma et al.

Summary: This study investigated the effect and mechanism of anemonin (ANE) on H2O2 induced degeneration of nucleus pulposus cells (NPCs). The results showed that ANE inhibited H2O2-induced inhibition of NPCs activity, oxidative stress, inflammation, and extracellular matrix degradation. These effects may be mediated through the inhibition of the NOX4/NF-kappa B pathway. Therefore, ANE might be a candidate drug for the treatment of intervertebral disk degeneration (IVDD).

Anemonin inhibits sepsis-induced acute kidney injury via mitigating inflammation and oxidative stress

Dan Liu et al.

Summary: In this study, the protective effects of the natural compound Anemonin (ANE) on sepsis-induced acute kidney injury (AKI) were investigated. ANE improved apoptosis, inflammatory response, and oxidative stress (OS) in renal tubular epithelial cells in a dose-dependent manner. ANE activated the AMPK/Sirt-1 pathway and attenuated AKI in a septic mouse model. These findings suggest that ANE has protective effects against AKI through repression of inflammation, OS, and cell apoptosis via activation of the AMPK/Sirt-1 pathway.

Anti-inflammatory effects of anemonin on acute ulcerative colitis via targeted regulation of protein kinase C-θ

Lu Jiang et al.

Summary: The study found that anemonin can improve body weight loss, shortened colon length, increased DAI, and induced pathological changes in the colon tissue of UC mice. Anemonin inhibits DSS-induced colon tissue inflammation and also attenuates LPS-induced cytokine production in HT-29 cells. The anti-inflammatory effects of anemonin may be mediated through targeting PKC-theta.

Evolving concepts in NAD+ metabolism

Claudia C. S. Chini et al.

Summary: NAD and NADP have diverse roles beyond co-factors in oxidation-reduction reactions, including signaling pathways, post-translational modifications, epigenetic changes, and RNA regulation. Recent studies have shown that NAD metabolism is highly dynamic with a short halflife in some tissues, and new concepts are emerging in the roles of NAD pathway metabolites in various disease states. Resolving open questions in NAD biology will lead to significant advancements in the field.
