Methyltransferase-like 3 leads to lung injury by up-regulation of interleukin 24 through N6-methyladenosine-dependent mRNA stability and translation efficiency in mice exposed to fine particulate matter 2.5

Xiang He et al.

Summary: This study reveals the significant role of m6A modification in lung injury caused by PM2.5 exposure. The findings suggest that PM2.5 exposure increases the total level of m6A modification and the expression of METTL3. This upregulation of METTL3 leads to increased expression and m6A modification of IL24, enhancing the stability of IL24 mRNA. Additionally, PM2.5 exposure also upregulates the expression of YTHDF1, which augments the translation efficiency of IL24 mRNA. Downregulation of Mettl3 reduces IL24 expression and improves pulmonary inflammation and mucus secretion in PM2.5-exposed mice.

Sestrin2 protects against cholestatic liver injury by inhibiting endoplasmic reticulum stress and NLRP3 inflammasome-mediated pyroptosis

Daewon Han et al.

Summary: Sestrin2 plays a crucial role in cholestatic liver injury, with its deficiency leading to heightened ER stress and severe liver damage. Additionally, Sestrin2 deficiency promotes cholestasis-induced hepatic pyroptosis.

Hsa_circ_0045714 induced by eupatilin has a potential to promote fracture healing

Yan Sun et al.

Summary: The study demonstrated that eupatilin can enhance the survival, proliferation, and migration of preosteoblasts, as well as promote the expression of fracture-related proteins. Moreover, it was found that hsa_circ_0045714 is increased in patients with hand and calcaneus fractures, and is involved in the therapeutic effects of eupatilin. These results suggest that eupatilin may be a promising candidate for fracture healing.

Sestrin2 is an endogenous antioxidant that improves contractile function in the heart during exposure to ischemia and reperfusion stress

Yunxia Liu et al.

Summary: Sestrin2 (Sesn2) is a stress-inducible protein that plays a critical role in responding to ischemic stress by reducing the generation of reactive oxygen species (ROS). Rescue of Sesn2 levels in Sesn2 KO hearts improved cardiac function, ameliorated ROS generation, and improved cardiomyocyte contractile function under ischemia and reperfusion. Overall, these findings highlight the role of Sesn2 as an endogenous antioxidant in maintaining intracellular redox homeostasis during ischemic stress.

Recent Updates on the Involvement of PI3K/AKT/mTOR Molecular Cascade in the Pathogenesis of Hyperproliferative Skin Disorders

Laura Mercurio et al.

Summary: PI3K represents a family of kinases that control multiple biological processes in mammalian cells and is associated with various skin pathologies. The dysregulation of PI3K/AKT/mTOR pathway contributes to uncontrolled cell proliferation and resistance to apoptosis in skin cancers, while also leading to enhanced proliferation, defective differentiation, and anti-apoptosis in psoriasis. The role of PI3K/AKT/mTOR in atopic dermatitis is less characterized, with recent evidence highlighting its function in epidermal barrier formation and stratification.

SESN2 protects against denervated muscle atrophy through unfolded protein response and mitophagy

Xiaofan Yang et al.

Summary: The study showed that after denervation of skeletal muscles, the stress-induced protein SESN2 was induced to regulate endoplasmic reticulum stress, mitochondrial dysfunction, and apoptosis, and its increase can protect against severe atrophy by inhibiting protein synthesis and promoting mitophagy.
