Allosteric quinoxaline-based inhibitors of the flavivirus NS2B/NS3 protease

Summary: Viruses from the Flavivirus genus, such as dengue virus (DENV) and Zika virus (ZIKV), infect millions of people worldwide and have no antiviral treatment. This study successfully identified quinoxaline-based allosteric inhibitors as a potential new drug-like scaffold to target the flavivirus NS2B/NS3 protease. Enzymatic assays, mutational analysis, and antiviral activity confirmed the therapeutic potential of these inhibitors.
