Development of 1-(2-aminophenyl)pyrrole-based amides acting as human topoisomerase I inhibitors

5,8-Dimethyl-9H-carbazole Derivatives Blocking hTopo I Activity and Actin Dynamics

Summary: Carbazoles have been extensively studied for their various biological properties, including their potential as antibacterial, antimalarial, antioxidant, antidiabetic, neuroprotective, and anticancer agents. This study focused on the anticancer activity of a series of carbazole derivatives against breast cancer cell lines. Compounds 3 and 4 showed the highest activity against the triple negative MDA-MB-231 cell line without affecting normal cells. Further investigations revealed that these compounds selectively inhibited human topoisomerase I and disrupted the actin system, leading to apoptosis. Therefore, compounds 3 and 4 hold promise for the development of multi-targeted therapy for triple negative breast cancer.

This review focuses on the combined application of irinotecan and doxorubicin, two extensively researched topoisomerase inhibitors, for colorectal and breast cancer therapy.

Design, synthesis of DNA-interactive 4-thiazolidinone-based indolo-/pyrroloazepinone conjugates as potential cytotoxic and topoisomerase I inhibitors

Summary: This study designed and synthesized a series of new small molecular entities for chemotherapy, among which conjugate 11g showed the strongest anti-cancer activity and high selectivity. Further evaluations demonstrated the good cytotoxicity and apoptosis-inducing ability of 11g and 12g. Molecular modeling and toxicity analysis provided further support for the drug-likeness and safety of these compounds. These findings are of great significance for cancer drug development.

Exploring the anticancer and antioxidant properties of Vicia faba L. pods extracts, a promising source of nutraceuticals

The extracts from broad bean pods were found to have good in vitro antioxidant and anticancer activities, especially against the melanoma cell line Sk-Mel-28. Furthermore, the ethanol extract was found to induce apoptosis in cancer cells by interfering with microtubule organization.

Deep scaffold hopping with multimodal transformer neural networks

Summary: Scaffold hopping is essential in modern medicinal chemistry for designing molecules with novel scaffolds but similar biological activities to known compounds. In this study, a supervised molecule-to-molecule translation approach was used to generate hopped molecules with similar 3D structures but different 2D structures. The trained DeepHop model successfully produced molecules with improved bioactivity and high 3D similarity compared to template molecules.
