Discovery of new 6-ureido/amidocoumarins as highly potent and selective inhibitors for the tumour-relevant carbonic anhydrases IX and XII

Summary: A series of 6-ureido/amidocoumarins were designed and synthesised to develop potent and isoform-selective carbonic anhydrase inhibitors. The target coumarins effectively inhibited tumour-related isoforms hCA IX and hCA XII, while not inhibiting cytosolic off-target isoforms hCA I and II. The most promising compound, 3,5-bis-trifluoromethylphenyl ureidocoumarin 5i, showed the best anticancer activity.

Tail-Approach-Based Design and Synthesis of Coumarin-Monoterpenes as Carbonic Anhydrase Inhibitors and Anticancer Agents

Summary: In this study, sixty novel coumarin-monoterpene compounds were synthesized and evaluated for their inhibitory effects on hCA isoforms and anticancer potentials. All synthesized molecules selectively inhibited CA IX and XII, with compounds 23 and 42 showing the strongest inhibitory activity against hCA IX. Cytotoxicity evaluation revealed that compounds 14 and 63 exhibited the highest cytotoxicity on MCF-7 cells, while compound 23 showed the strongest cytotoxic effect on both PC-3 and HT-29 cell lines. Furthermore, compounds 14, 23, and 66 showed a reduction in CA IX and CA XII protein expression and increased apoptosis in different cell lines. The modeling studies demonstrated that only the open coumarin form of the compounds could interact directly with the active-site Zn2+ ion.

Synthesis and biological evaluation of new 3-substituted coumarin derivatives as selective inhibitors of human carbonic anhydrase IX and XII

Summary: Carbonic anhydrase isoforms IX and XII play a crucial role in regulating pH in hypoxic tumors and promoting the metastasis of solid tumors. Selective inhibitors targeting these isoforms can reduce their activity, providing an anti-tumor and anti-metastatic mechanism. Coumarin-based derivatives are selective inhibitors of CA isoforms IX and XII. In this study, new 3-substituted coumarin derivatives were designed and synthesized, and their inhibitory activity against various carbonic anhydrase isoforms was evaluated. Tertiary sulphonamide derivative 6c showed selective inhibition against CA IX, while carbothioamides 7c, 7b, and oxime ether derivative 20a exhibited good inhibition against both CA IX and CA XII. The binding mode was predicted and validated using molecular docking and dynamic simulations.

Coumarin and Piperazine Conjugates as Selective Inhibitors of the Tumor-associated Carbonic Anhydrase IX and XII Isoforms

Summary: The objective of this study was to synthesize potent and selective inhibitors of tumor-associated carbonic anhydrases (CA IX and XII). A library of 17 coumarin derivatives was designed, synthesized, and evaluated for their inhibitory effects and selectivity towards relevant CA isoforms I, II, IX, and XII. The compounds showed activity against tumor-associated isoforms IX and XII, with the most active compound having a Ki of 229 nM for hCA IX and 294.2 nM for hCA XII. Additionally, two compounds exhibited exquisite selectivity towards off-target isoforms I and II, and were approximately 20-fold more selective towards hCA IX than XII. Molecular modeling techniques provided insights into the observed selectivity.

2-Aminobenzoxazole-appended coumarins as potent and selective inhibitors of tumour-associated carbonic anhydrases

Summary: In this study, a small library of 2-aminobenzoxazole-appended coumarins was designed, synthesized, and evaluated as novel inhibitors of tumour-related CA IX and XII. The structure-activity relationships were established through modifications on substituents and linker length. The compounds exhibited good selectivity towards CA IX and XII and the introduction of a chlorine atom on the coumarin moiety showed the strongest antiproliferative activity.

Coumarins effectively inhibit bacterial α-carbonic anhydrases

Summary: Coumarins were shown to inhibit bacterial alpha-carbonic anhydrases from Neisseria gonorrhoeae and Vibrio cholerae, with the nature and position of substituents in the coumarin ring strongly influencing the inhibitory efficacy. In comparison, human CA isoforms hCA I and II were less prone to inhibition by these compounds, indicating potential for coumarin bacterial CA inhibitors with selectivity for bacterial over human isoforms.

Binding site comparison for coumarin inhibitors and amine/amino acid activators of human carbonic anhydrases

Summary: This study presents the first structural analysis comparing two opposite classes of modulators binding to the target carbonic anhydrases, showing that coumarin derivatives act as prodrug CA inhibitors while CA activators belonging to the amine and amino acid types enhance CA activity by increasing the efficiency of the rate-determining proton shuttling step in the CA catalytic cycle. The analysis of crystallographic data reveals that both types of CA modulators bind in the same region of the enzyme active site, interacting with superimposable amino acid residues and involving a plethora of water molecules in the formation of adducts. Additionally, the study highlights the importance of certain chemical groups in compound structure to produce an activating rather than inhibitory action, with multiple nitrogen- and oxygen-based moieties capable of shuttling protons or forming extended H-bond networks nearby the proton shuttle residue. This unique finding showcases the significant pharmacological implications of having an identical binding site for both inhibitors and activators in enzymes.
