Novel findings about the mode of action of the antifungal protein PeAfpA against Saccharomyces cerevisiae

The Penicillium digitatum antifungal protein PdAfpB shows high activity against mycobiota involved in sliced bread spoilage

Summary: This study aimed to isolate and identify fungal species involved in sliced bread spoilage, and to evaluate their susceptibility to antifungal proteins of fungal origin. The study found that Penicillium species were the predominant mycobiota, with P. roqueforti, P. brevicompactum, P. chrysogenum and P. crustosum being the most abundant species. PdAfpB and PeAfpA were the most potent proteins in inhibiting fungal growth, while PeAfpC was the least active protein.

Rationally designed antifungal protein chimeras reveal new insights into structure-activity relationship

Summary: Antifungal proteins (AFPs) produced by Penicillium expansum showed different antifungal profiles and fruit protection effects. By solving the crystal structure of PeAfpB and designing five PeAfpA::PeAfpB chimeras, researchers found that the man-noproteins determined the protein interaction with the cell wall and its antifungal activity, while the binding to membrane phospholipids did not directly correlate with activity. This work provides insights into the feasibility of modifying protein specificity through rational design of chimeras with biotechnological applicability.

Impact of the antifungal protein PgAFP on the proteome and patulin production of Penicillium expansum on apple-based medium

Summary: The antifungal protein PgAFP may lead to overproduction of the mycotoxin patulin in Penicillium expansum on apples. Proteomic analysis showed an increase in proteins involved in patulin biosynthesis and a decrease in proteins related to pathogenicity and virulence in PgAFP-treated P. expansum. Additionally, proteins associated with oxidative stress were found in higher abundance, indicating a response to oxidative stress elicited by PgAFP.

In this study, small cationic antifungal proteins from filamentous fungi show promise as next-generation antifungals for topical application, demonstrating efficacy against cutaneous C. albicans infection in a 3D skin model.

Differential susceptibility of mycotoxin-producing fungi to distinct antifungal proteins (AFPs)
