Recent structural studies on prenyltransferases shed light on their catalytic mechanisms and key residues, but the control of regioselectivity and chemoselectivity remains a challenge. Site-directed mutagenesis has been successfully utilized to modify prenyl donor specificity and switch the regioselectivity of prenyltransferases, showing potential for production of novel prenylated compounds.

Summary: In this study, successful prenylation or geranylation of biflavonoids using fungal prenyltransferase CdpC3PT and its mutants was reported. F253 was identified as a key residue related to donor selectivity, enabling the precise switching from utilizing DMAPP to GPP at the same C-3''' site of biflavonoids. Furthermore, residue W181 was discovered to generally increase prenylation activity towards biflavonoids.

Recent attention has been focused on cyclodipeptide synthase-associated P(450)s, as they can catalyze a wider range of reactions beyond oxidation.

Global Genome Mining Reveals a Cytochrome P450-Catalyzed Cyclization of Crownlike Cyclodipeptides with Neuroprotective Activity

Summary: By conducting global genome mining of bacterial genomes, 829 cyclodipeptide biosynthesis gene clusters containing a cytochrome P450 gene were identified. Heterologous expression of these gene clusters led to the discovery of two novel crownlike cyclodipeptides, cyctetryptomycin A and B, which exhibit complex macrocycle structures and neuroprotective activity. Two cytochrome P450 enzymes catalyze sequential reactions resulting in the cyclization of diketopiperazine dimers within the gene clusters.
