Pyrimidine inhibitors synergize with nucleoside analogues to block SARS-CoV-2

David C. Schultz et al.

Summary: This study screened and validated 122 drugs with antiviral activity and selectivity against SARS-CoV-2 out of approximately 18,000 drugs. Among them are the approved antivirals remdesivir and molnupiravir. The study also found that combining pyrimidine biosynthesis inhibitors with antiviral nucleoside analogues synergistically inhibits SARS-CoV-2 infection.

Inhibitors of Nucleotide Biosynthesis as Candidates for a Wide Spectrum of Antiviral Chemotherapy

Claudia Soledad Sepulveda et al.

Summary: Host-targeted antivirals (HTA) are an interesting strategy to combat viruses by inhibiting cellular molecules or pathways involved in virus multiplication. Inhibitors of nucleotide metabolism, particularly the rate-limiting enzymes DHODH and IMPDH, have shown promising antiviral properties against a broad spectrum of pathogenic viruses.

Summary: The need for new drug treatments for COVID-19, caused by SARS-CoV-2, is highlighted due to the virus's continued evolution. One potential strategy is the combination of BRQ and DPY to enhance depletion of the cellular pyrimidine nucleotide pool and show antiviral activity against SARS-CoV-2 variants.

Summary: Infections caused by microscopic viruses can lead to chronic illnesses like HIV/AIDS, flu, etc. Developing new and superior antiviral drugs is crucial, but the drugs can be toxic to human cells and viruses can develop resistance. Combination drugs targeting viral replication are common, but innovative strategies are needed to combat emerging viruses like Ebola and coronaviruses.

Broad-Spectrum Antiviral Strategies and Nucleoside Analogues

Robert J. Geraghty et al.

Summary: The emergence or re-emergence of viruses with epidemic and pandemic potential poses significant human health threats, and although vaccines are key in controlling virus spread, their development may face obstacles such as safety concerns. Developing broad-spectrum antiviral molecules, particularly nucleoside analogues, for a fast response in emergency situations is crucial in combating outbreak crises.
