Integration of Mendelian randomisation and systems biology models to identify novel blood-based biomarkers for stroke

Summary: This study aimed to identify putative functional causal gene biomarkers of stroke risk using a summary-based Mendelian randomisation (SMR) approach. Through integrative analysis, 45 candidate biomarker genes were identified that were pleiotropically or potentially causally associated with stroke risk. Among them, 10 genes exhibited differential expression in blood transcriptomics data from stroke and healthy individuals. This study provides insights into the influence of DNA methylation on the expression of genes linked to stroke risk.

Most genes show cis-eQTL effects, while distal trans-eQTL effects have lower replication rates. However, single-cell RNA-seq data prioritize intracellular trans-eQTL effects mediated by transcription factors. Additionally, a subset of genes correlated with polygenic scores for various phenotypes may serve as potential drivers for those traits.

Pleiotropy robust methods for multivariable Mendelian randomization

Summary: Mendelian randomization is a powerful method for inferring causal effects from observational data, but the presence of pleiotropy in genetic variants remains a challenge for accurate estimation. While there are various pleiotropy robust methods available for studying single risk factors, there are limited options for the multivariable setting with multiple risk factors. This study introduces three new methods and evaluates their performance in simulation studies, demonstrating varying levels of effectiveness depending on the level of pleiotropy present.
