A Universal DNA Aptamer that Recognizes Spike Proteins of Diverse SARS-CoV-2 Variants of Concern

Zijie Zhang et al.

Summary: A unique DNA aptamer, MSA52, has been identified in this study, which exhibits high affinity for the spike proteins of wildtype SARS-CoV-2 and multiple variants. This discovery demonstrates the potential of generating aptamers with high affinity for multiple variants of a single protein, including emerging variants, which makes it a promising tool for molecular recognition of rapidly evolving targets, such as those found in SARS-CoV-2.

Jiuxing Li et al.

Summary: Researchers have discovered a new homotrimeric aptamer called TMSA52, which has excellent binding affinity and can bind to various SARS-CoV-2 spike protein variants. The study also showed that using TMSA52 as a probe in an enzyme-linked aptamer binding assay can sensitively detect different SARS-CoV-2 variants in human saliva samples.

Neutralizing Aptamers Block S/RBD-ACE2 Interactions and Prevent Host Cell Infection

Xiaohui Liu et al.

Summary: The receptor-binding domain of the SARS-CoV-2 spike protein is crucial for mediating virus entry into human cells, making it an ideal target for blocking infection. Using aptamers specifically targeting the S/RBD, researchers were able to effectively prevent virus binding to host cells and neutralize viral particles expressing the S protein, demonstrating a promising COVID-19 therapeutic strategy.

Diverse high-affinity DNA aptamers for wild-type and B.1.1.7 SARS-CoV-2 spike proteins from a pre-structured DNA library

Jiuxing Li et al.

Summary: In vitro selection experiments identified DNA aptamers, MSA1 and MSA5, with strong binding affinity to the S1 subunit of the SARS-CoV-2 spike protein. These aptamers demonstrated comparable affinities for various forms of S1 protein and also recognized the pseudotyped lentivirus of SARS-CoV-2, highlighting their potential as diagnostic tools for COVID-19 detection. The development of a colorimetric sandwich assay using MSA1 further confirmed the utility of these aptamers in detecting the virus in saliva samples.

Direct detection of human adenovirus or SARS-CoV-2 with ability to inform infectivity using DNA aptamer-nanopore sensors

Ana S. Peinetti et al.

Summary: A new method has been developed for direct detection and differentiation of infectious viruses, including human adenovirus and SARS-CoV-2, from noninfectious ones without the need for sample pretreatment. This method utilizes DNA aptamers to bind intact infectious viruses and incorporates them into a solid-state nanopore for enhanced sensitivity. Applications of this sensor have been demonstrated in various types of samples, making it generally applicable for detecting emerging viruses of environmental and public health concern.
