Digital Microfluidics and Magnetic Bead-Based Intact Proteoform Elution for Quantitative Top-down Nanoproteomics of Single C. elegans Nematodes

Jan Leipert et al.

Summary: Researchers have developed a sample preparation method based on protein aggregation and acidic elution, allowing for sensitive protein identification analysis of single nematodes on a digital microfluidics device. Compared to in-tube sample preparation, this method increases proteoform identifications by 46%. Label-free quantification also reveals changes in proteoform abundance that cannot be distinguished by bottom-up proteomics. This workflow facilitates proteoform-focused analysis on limited availability samples.

2′-O-Methyl modified guide RNA promotes the single nucleotide polymorphism (SNP) discrimination ability of CRISPR-Cas12a systems

Yuqing Ke et al.

Summary: In this study, we propose a strategy to enhance the specificity of the Cas12a system by using 2'-O-methyl modified guide RNA (gRNA), and we validate the effectiveness of this strategy through experiments. Our results show that the 2'-OMe modified gRNAs can significantly improve the specificity of Cas12a in discriminating single-base mutations, and molecular docking simulations reveal the mechanism of this enhancement.

Streamlined single-cell proteomics by an integrated microfluidic chip and data-independent acquisition mass spectrometry

Sofani Tafesse Gebreyesus et al.

Summary: Single-cell proteomics can reveal cellular heterogeneity and functional networks underlying biological processes. We present a streamlined workflow combining microfluidic chips and data-independent acquisition mass spectrometry for single-cell proteomic analysis. This workflow allows for efficient isolation, counting, imaging, and sample processing of cells in a single device, enabling the analysis of protein groups in individual mammalian cells. The chip-DIA workflow provides a basis for advanced single-cell proteomics applications.

Unbiased spatial proteomics with single-cell resolution in tissues

Andreas Mund et al.

Summary: Mass spectrometry-based proteomics is a powerful technology for quantifying proteins in cells or tissues. Single-cell transcriptomes are influenced by stochastic noise due to low transcript numbers, whereas single-cell proteomes appear to be more complete. With the combination of imaging and spatial transcriptomics, the spatial organization of cells in tissues can be studied alongside ultra-sensitive proteomics.

Enhanced Competition at the Nano-Bio Interface Enables Comprehensive Characterization of Protein Corona Dynamics and Deep Coverage of Proteomes

Shadi Ferdosi et al.

Summary: This study introduces the formation of a selective and reproducible protein corona at the interface of engineered nanoparticles (NPs) and biofluid, and models the dynamics of protein corona formation. Adjusting the protein to NP-surface ratio significantly affects the composition of protein corona and enhances the depth and precision of proteomic studies. Moreover, the ratio of protein to NP-surface plays a crucial role in determining the fate of nanomaterials in biomedical applications.

Hairpin-Spacer crRNA-Enhanced CRISPR/Cas13a System Promotes the Specificity of Single Nucleotide Polymorphism (SNP) Identification

Yuqing Ke et al.

Summary: This study demonstrates the potential of hairpin-spacer crRNAs in improving specificity and sensitivity of the CRISPR/Cas13a system. The high-specificity RNA editing tool establishes a new approach for rational crRNA design, offering significant improvements in the Cas13a system. Theoretical simulations elucidate the mechanism of Cas13a/hs-crRNA/target RNA interaction, further enhancing our understanding of the system's activation.
