Virus-like Particle Vaccines and Platforms for Vaccine Development

Milad Kheirvari et al.

Summary: Virus-like particles (VLPs) have gained significant interest as vaccines. They have been approved for protection against various infectious agents and can also serve as platforms for displaying foreign peptide antigens. This review summarizes approved and under-development VLP vaccines for human and veterinary use, as well as chimeric VLP vaccines tested in pre-clinical studies. It concludes with the advantages of VLP-based vaccines over conventional approaches.

Structure Guided Design of Bacteriophage Qβ Mutants as Next Generation Carriers for Conjugate Vaccines

Suttipun Sungsuwan et al.

Summary: Vaccines are critical tools for treating and preventing diseases. However, there are few available carriers for effective conjugate vaccines, and current protein carriers often induce high levels of antibodies against themselves, interfering with immune responses. Researchers have obtained the crystal structure of a promising protein carrier called bacteriophage Qfl virus-like particle (mQfl), and designed mutants of mQfl that can elicit strong immune responses against tumor antigens while producing lower levels of anti-carrier antibodies. In mouse models, immunization with mQfl-TACA conjugates resulted in a significant reduction in tumor growth compared to unimmunized mice. Moreover, mQfl has shown potential as a carrier for developing vaccines against COVID-19, inducing high levels of IgG antibodies against peptide antigens from the SARS-CoV-2 virus. Structure-based rational design is a powerful strategy for developing new vaccine carriers.

Targeting Tumor Glycans for Cancer Therapy: Successes, Limitations, and Perspectives

Nora Berois et al.

Summary: Aberrant glycosylation is a common feature of cancer and can lead to changes that affect tumor behavior. Glycans can serve as a source of new clinical biomarkers, providing specific targets for therapeutic intervention. Different mechanisms of aberrant glycosylation lead to the formation of tumor-associated carbohydrate antigens (TACAs) suitable for selective cancer-targeting therapy.
