Structural Basis of Sirtuin 6-Catalyzed Nucleosome Deacetylation

Summary: The reversible acetylation of histone lysine residues is controlled by the action of acetyltransferases and deacetylases (HDACs), which regulate chromatin structure and gene expression. One member of the HDAC family, Sirt6, efficiently deacetylates several histone H3 acetylation sites on nucleosomes. The structure of Sirt6 bound to a nucleosome reveals its extensive interactions with H2A/H2B acidic patch and nucleosomal DNA, providing insights into how HDACs target and regulate chromatin.

Cryo-EM structure of the human Sirtuin 6-nucleosome complex

Summary: SIRT6 is a versatile protein deacetylase/deacylase that is targeted by small molecules for longevity and cancer. Its ability to remove acetyl groups from histone H3 in chromatin and its nucleosomal substrate preference remain unknown. Our cryo-electron microscopy structure reveals that SIRT6 pries DNA from the nucleosomal entry-exit site and exposes the histone H3 N-terminal helix, while its zinc-binding domain binds to the histone acidic patch. Additionally, SIRT6 interacts inhibitory with the C-terminal tail of histone H2A. These findings provide insights into the deacetylation of both H3 K9 and H3 K56 by SIRT6.

Potent Activation of NAD+-Dependent Deacetylase Sirt7 by Nucleosome Binding

Summary: Sirtuin-7 (Sirt7) is an important protein enzyme with a wide range of biological functions and potential therapeutic value. This study reveals that Sirt7 preferentially interacts with nucleosomes and shows the maximum enzymatic activity only when bound to nucleosomes. The catalytic activity of Sirt7 is limited by the rate of dissociation from deacetylated nucleosomes, as determined by the analysis of its catalytic mechanism.
