High DNA stainability (HDS) should not be recommended as a marker for the detection of sperm DNA damage

Jin-Chun Lu

Summary: A marker called high DNA stainability (HDS) is used to detect sperm DNA damage. However, there is controversy surrounding its definition and related research. Some studies reported an increase in the proportion of sperm with HDS with age, which correlated with poor outcomes in assisted reproductive technology. However, recent research found a linear decrease in the marker of HDS with age, which contradicts previous findings. After extensive research, it is concluded that the establishment of HDS in detecting sperm DNA damage lacks theoretical basis and evidence-based medicine support, making it inappropriate to use HDS as a marker.

A nomogram to assist blastocyst selection in vitrified-warmed embryo transfer cycles

Feng Xiong et al.

Summary: The study aims to establish a mathematical model to rank the order of blastocysts and assist selection in vitrified-thawed embryo transfer cycles. The model utilizes LASSO regression to identify critical factors for live birth (LB) and creates a nomogram to convert their effects into measurable scores. The model's performance is assessed using receiver operating characteristic and calibration curves, with internal testing conducted in a separate set. The nomogram model provides guidance for embryo selection, helping optimize live birth rates.

To use or not to use propensity score matching?

Jixian Wang

Summary: The text discusses the application of propensity score matching (PSM) in observational studies and its pros and cons. The authors argue that the right question should be when and how to use PSM, rather than simply choosing whether to use it or not, and provide corresponding suggestions.

Clinical implications of sperm DNA damage in IVF and ICSI: updated systematic review and meta-analysis

Jordi Ribas-Maynou et al.

Summary: A systematic review and meta-analysis of studies on IVF and ICSI treatments revealed a significant negative impact of sperm DNA damage on implantation and pregnancy rates. Routine sperm DNA testing is justified to improve IVF outcomes and assist in treatment selection for couples. Further studies are needed to draw more precise conclusions, especially in the context of ICSI treatments.

Elevated sperm DNA fragmentation does not predict recurrent implantation failure

Jordan C. Best et al.

Summary: The study aimed to investigate whether sperm DNA fragmentation (DFI%) and high DNA stainability (HDS%) evaluated by sperm chromatin structure assay (SCSA) can predict recurrent implantation failure (RIF) or pregnancy rate. After analyzing a total of 1,216 frozen embryo transfer (FET) cycles from 386 couples, it was found that DFI% and HDS% were not associated with RIF or IVF success, suggesting that sperm DNA fragmentation does not predict RIF or pregnancy rate.

Decreased DNA Repair Ability: A Mechanism for Low Early Embryonic Development Potential of Oocytes From Overweight Patients After Fertilization in IVF Cycles

Hui Li et al.

Summary: This study aimed to assess the impact of female BMI on DNA repair ability in oocytes after fertilization. It found that overweight women had lower embryo quality and reproductive outcomes when fertilized with sperm with high DNA fragmentation. The decreased DNA repair activity in oocytes may explain the low early development potential of embryos in overweight patients during in vitro fertilization cycles.
