Directly imaging emergence of phase separation in peroxidized lipid membranes

Miguel Paez-Perez et al.

Summary: Lipid peroxidation plays a crucial role in cell signaling, diseases, and therapy. The authors employed advanced microscopy, X-ray diffraction, and molecular simulations to directly measure the changes in bilayer structure and viscoelastic properties upon peroxidation of lipid membranes.

HydroFlipper membrane tension probes: imaging membrane hydration and mechanical compression simultaneously in living cells

Jose Garcia-Calvo et al.

Summary: HydroFlippers are fluorescent membrane tension probes that report on both membrane compression and hydration. Different cellular membranes show varied responses to tension-induced decompression and hydration. The mechanical compression is independent of the nature of the membrane of interest (MOI), while the response to hydration depends on the intrinsic order of the MOI.

LAURDAN since Weber: The Quest for Visualizing Membrane Heterogeneity

German Gunther et al.

Summary: Chemists studying membrane bilayer heterogeneity may encounter phase segregation in artificial bilayers and small sterol and sphingolipid-enriched dynamic domains in biological bilayers. Techniques using fluorescent probes and microscopy can assess membrane heterogeneity at different temporal and spatial levels, providing valuable information about the physical-chemical aspects of the membrane.

EPA and DHA differentially modulate membrane elasticity in the presence of cholesterol

Miranda L. Jacobs et al.

Summary: Polyunsaturated fatty acids (PUFAs) are hypothesized to modulate protein activity by indirectly altering membrane physical properties, with the membrane area expansion modulus (K-a) being a significant controller of channel activity. PUFAs generally decrease the apparent K-a (K-app) of membranes, and in membranes with high levels of cholesterol, docosahexaenoic acid (DHA) has a greater impact on reducing K-app compared to eicosapentaenoic acid (EPA). This study suggests that membrane phase behavior and the distribution of membrane-elasticizing amphiphiles affect the ability of a membrane to stretch.

Effect of Cholesterol Versus Ergosterol on DPPC Bilayer Properties: Insights from Atomistic Simulations

Azadeh Alavizargar et al.

Summary: Sterols, particularly cholesterol, play a major role in the organization of biological membranes and the formation of liquid ordered domains in lipid mixtures. Molecular dynamics simulations show that cholesterol has a stronger impact on phospholipid properties compared to ergosterol, likely due to its higher planarity in the ring system.

Imaging non-classical mechanical responses of lipid membranes using molecular rotors

Miguel Paez-Perez et al.

Summary: Lipid packing in cellular membranes affects membrane tension and microviscosity, influencing cellular adaptation, homeostasis, and disease. The relationship between membrane tension and microviscosity is strongly influenced by the lipid composition of the bilayer, with different membrane structures exhibiting varying responses to tension.
