Dual Signal Amplification by Urease Catalysis and Silver Nanoparticles for Ultrasensitive Colorimetric Detection of Nucleic Acids

An ultrasensitive ELISA to assay femtomolar level SARS-CoV-2 antigen based on specific peptide and tyramine signal amplification

Summary: The rapid spread of SARS-CoV-2 has led to catastrophic public healthcare issues and economic crises worldwide. A phage expression of WNLDLSQWLPPM peptide specific to SARS-CoV-2 spike protein (SP) was screened and a peptide-antigen-antibody ELISA (p-ELISA) with tyramine signal amplification (TSA) was developed, which showed an ultrasensitive detection limit of 14 fM and 2.8 fM for first TSA and secondary TSA, respectively. The proposed p-ELISA method with TSA could detect SARS-CoV-2 pseudoviruses down to 10 and 3 TCID50/mL in healthy nasal swab samples. This method is expected to be a promising tool for rapid detection of SARS-CoV-2 antigen and control of the infectious disease.

Summary: The COVID-19 pandemic has caused a global crisis, and detecting SARS-CoV-2 with high sensitivity is crucial for diagnosis and control. In this study, a specific phage peptide targeting the SARS-CoV-2 S1 protein was identified using a phage display library. The identified peptide showed good specificity and was used to establish a sandwiched phage-based enzyme-linked chemiluminescence immunoassay (ELCLIA) with high sensitivity. This method can detect as low as 78 pg/mL SARS-CoV-2 S1.

A paper-based assay for the colorimetric detection of SARS-CoV-2 variants at single-nucleotide resolution

Nanozyme-strip for rapid and ultrasensitive nucleic acid detection of SARS-CoV-2

Summary: The COVID-19 pandemic has led to a high demand for rapid and sensitive detection of SARS-CoV-2. In this study, we developed a method combining RPA and FeS2 nanozyme strips for nucleic acid amplification and colorimetric signal enhancement. The FeS2 nanozymes exhibited high peroxidase-like activity, enabling colorimetric signal amplification using a non-toxic substrate. The method showed a limit of detection for SARS-CoV-2 close to that of RT-PCR, and demonstrated high agreement with previous RT-PCR results for clinical samples. Our findings suggest that nanozyme-based nucleic acid detection has great potential in the development of POCT diagnosis for COVID-19 and other viral infections.

A pH-responsive bioassay for sensitive colorimetric detection of adenosine triphosphate based on switchable DNA aptamer and metal ion-urease interactions

DNAzyme Amplified Aptasensing Platform for Ochratoxin A Detection Using a Personal Glucose Meter

By conjugating biotin-labeled aptamer probe and substrate strand on magnetic beads, and hybridizing DNAzyme strand to block substrate cleavage, specific binding of small-molecular targets can be detected and quantified effectively through an iterative signal amplification reaction. This platform enables the detection of toxins such as ochratoxin with a wide linear detection range, low detection limit, and the potential for portable, at-home analysis of small molecules using a glucose meter.
