Metabolic modulation of immune checkpoints and novel therapeutic strategies in cancer

Yi Wang et al.

Summary: Cytotoxic T-lymphocyte-associated antigen-4 (CTLA-4) or programmed death-1 (PD-1)/programmed death-ligand 1 (PD-L1)-based immune checkpoint inhibitors (ICIs) have shown significant improvements in overall survival of certain cancer patients, but resistance to immunotherapy remains a challenge. This article examines the metabolic modulation of tumor immunity and immune checkpoints in the tumor microenvironment (TME) and discusses potential metabolism-based therapeutic strategies. Understanding the link between tumor metabolism and immunotherapy is crucial for cancer treatment and research.

Three-Dimensional Mass Spectrometry Imaging Reveals Distributions of Lipids and the Drug Metabolite Associated with the Enhanced Growth of Colon Cancer Cell Spheroids Treated with Triclosan

Peisi Xie et al.

Summary: The application of mass spectrometry imaging (MSI) in studying the response of cancer cell spheroids (CCS) to exogenous molecules has been important. In this study, a 3D MSI method was used to differentiate the distribution of triclosan sulfate and endogenous lipids in colon CCS. The results showed that triclosan sulfate accumulated gradually in CCS from the periphery to the core region. Spatial lipidomics analysis revealed different distributions of upregulated lipid species in different regions of CCS, providing insights into the growth mechanism of CCS treated with triclosan.

Berberine treats atherosclerosis via a vitamine-like effect down-regulating Choline-TMA-TMAO production pathway in gut microbiota

Shu-Rong Ma et al.

Summary: This study found that berberine can treat atherosclerosis by reducing trimethylamine-N-oxide (TMAO) production in the gut microbiota. The results showed that berberine interacts with enzymes in the gut microbiota to lower TMAO biosynthesis, resulting in decreased TMAO levels in the blood and interrupted plaque formation in blood vessels. Clinical trials also demonstrated that oral berberine can decrease plaque score and reduce TMA and TMAO levels in atherosclerotic patients.

Lipid signalling enforces functional specialization of Treg cells in tumours

Seon Ah Lim et al.

Summary: Regulatory T cells play a crucial role in immune tolerance and immunosuppression in the tumor microenvironment. Inhibiting lipid synthesis dependent on SREBPs in T-reg cells can enhance antitumor immune responses without autoimmune toxicity, and deletion of SCAP in these cells inhibits tumor growth and enhances immunotherapy. SCAP and SREBPs signaling regulate lipid synthesis and inhibitory receptor signaling in T-reg cells, pointing to new ways of targeting these cells for cancer therapy.
